摘要
为探究鱼腥草桔蓝口服液治疗家禽呼吸道疾病的作用机制,本研究通过检索鱼腥草桔蓝口服液的化学成分及作用靶点,构建药物-靶点网络图和交集靶点蛋白质(PPI)相互作用网络图,并对交集靶点进行富集分析;通过分子对接技术对药物活性成分和核心靶点进行分子对接验证。结果显示,鱼腥草桔蓝口服液在槲皮素、山奈酚和β-谷甾醇等活性成分的作用下,对白细胞介素-6(IL-6)、B淋巴细胞瘤-2(BCL2)、信号转导和转录激活因子3(STAT3)等关键靶点产生影响;GO功能富集获得58条生物过程条目、4条细胞组成条目、15条分子功能条目;KEGG通路分析显示,交集基因主要作用于C型凝集素受体信号通路、FoxO信号通路、MAPK信号通路和VEGF信号通路等多条信号通路,对家禽呼吸道疾病进行治疗。分子对接结果显示,药物关键成分与核心蛋白靶点有较好的结合性。结果表明鱼腥草桔蓝口服液可通过多成分、多靶点、多通路的机制治疗家禽呼吸道疾病。
To investigate the action mechanism of Yuxingcao Jielan oral liquid on the treatment of respiratory diseases in poultry.In this study,the chemical composition and action targets of Yuxingcao Jielan oral liquid were searched,a drug-target network map and an intersecting target protein(PPI)interaction network map were constructed.The enrichment of intersecting targets was analyzed.The molecular docking validation of drug active ingredients and core targets were analyzed by molecular docking technology.The results showed that Yuxingcao Jielan oral liquid,in the presence of active ingredients such as quercetin,kaempferol andβ-sitosterol,affected key targets such as interleukin-6(IL-6),B-cell lymphoma-2(BCL2),signal transducer and activator of transcription 3(STAT3).GO functional enrichment yielded 58 biological process entries,4 cellular composition entries,and 15 molecular function entries.KEGG pathway analysis revealed that the intersectinggenes mainly acted on multiple signaling pathways such as C-type lectin receptor signaling pathway,FoxO signaling pathway,MAPK signaling pathway and VEGF signaling pathway to treat poultry respiratory diseases.The molecular docking results indicated good binding of the key components of the drug to the core protein target.The results indicated that Yuxingcao Jielan oral liquid could treat poultry respiratory diseases through a multi-component,multi-target and multi-pathway mechanism.
