紫朱软膏对糖尿病溃疡小鼠创面炎症反应及上皮-间充质转化的影响

Effect of Zizhu ointment on wound inflammation and epithelial-mesenchymal transformation of diabetic ulcer

目的:观察紫朱软膏对糖尿病溃疡小鼠创面炎症反应及上皮-间充质转化的影响。方法:采用C57B/L小鼠分别制备为普通溃疡模型和糖尿病溃疡模型,分为对照组、模型组、紫朱软膏组,连续干预14 d,观察小鼠创面愈合情况及愈合率。干预后采用qRT-PCR检测局部溃疡组织炎症相关因子白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、一氧化氮合酶(iNOS)、白介素-4(IL-4)、白介素-10(IL-10)的表达情况,采用Western blot、qRT-PCR方法检测上皮-间充质转化相关细胞因子锌指转录因子(Snail)、α-平滑肌肌动蛋白(α-SMA)、上皮细胞钙黏蛋白(E-cadherin)、β-连环蛋白(β-catenin)表达情况。结果:干预11、14 d,紫朱软膏组小鼠溃疡创面愈合率高于模型组(均P<0.01)。与模型组比较,紫朱软膏组小鼠溃疡创面组织IL-6、TNF-α、iNOS RNA表达降低,IL-4、IL-10 RNA表达升高(均P<0.05)。与模型组比较,紫朱软膏组小鼠溃疡创面组织snail、β-catenin、α-SMA表达上调,E-cadherin表达下调(均P<0.05)。结论:紫朱软膏可能通过减轻炎症反应,诱导上皮-间充质转化,促进糖尿病溃疡愈合。

Objective:To observe the effect of Zizhu ointment on wound inflammation and epithelium-mesenchymal transformation in diabetic ulcer mice.Methods:Common ulcer model and diabetic ulcer model were prepared by C57B/L mice,which were divided into control group,model group and Zizhu ointment group.The wound healing and healing rate were observed after continuous intervention for 14 days.After intervention,the expression of inflammation-related factors IL-6,TNF-α,iNOS,IL-4,IL-10 in local ulcer tissues was detected by qRT-PCR.The expression of epithelial-mesenchymal transformation related cytokines Snail,α-SMA,E-cadherin,β-catenin was detected by Western blot and qRT-PCR.Results:After 11 and 14 days of intervention,the healing rate of ulcer wound in Zizhu ointment group was higher than that in model group(all P<0.01).Compared with model group,the expression of IL-6,TNF-αand iNOS RNA in ulcer wound tissue of mice in Zizhu ointment group decreased,while the expression of IL-4 and IL-10 RNA increased(all P<0.05).Compared with model group,the snail,β-catenin andα-SMA expressions in ulcer wound tissue of mice in Zizhu ointment group were up-regulated,and E-cadherin expressions were down-regulated(all P<0.05).Conclusion:Zizhu ointment may promote the healing of diabetic ulcer by reducing inflammatory response and inducing epithelial-mesenchymal transformation.
