水解物对CHO细胞培养工艺的影响

摘要 目的对比3种水解物对CHO悬浮培养细胞生长的影响,以及对细胞分泌表达的抗体质量的影响。方法应用3种不同来源的水解物,采用摇瓶培养CHO细胞,观察培养过程中细胞密度、活性和代谢产物的变化,以及抗体质量的影响。结果水解物对CHO生长无显著促进作用、但可大幅度提高抗体产量;对抗体糖型无显著影响,可显著影响抗体电荷分布。结论3种水解物中,Preteose peptone和Phytone peptone对CHO有很好的促进作用。但Preteose peptone含有动物然产分,随着各国药监对动物来源成分监管制度的严格,将进一步限制Preteose peptone的应用,因此Phytone peptone具有优势。 Objective This study aims to assess the impact of three distinct hydrolysates on CHO suspension cell culture,specifically focusing on cell density,titer,and product quality.Methods To conduct this investigation,three different hydrolysates were introduced as medium supplements in a 1L shaker for cell culture.Daily records were maintained for cell density,viability,and titer,and product quality was assessed post-harvest.Results The results indicate that all three hydrolysates serve as valuable supplements to enhance productivity,albeit without a significant impact on CHO cell growth.It was observed that hydrolysates do not influence antibody glycan patterns,but they can alter the distribution pattern of antibody charge.Conclusions Among the 3 hydrolysates,Preteose peptone and Phytone peptone have good promotion effect on CHO.However,Preteose peptone contains animal-derived ingredients,which will further limit the application of Preteose peptone as the regulatory system of animal-derived ingredients becomes stricter in various countries'pharmacovigilance,thus Phytone peptone has an advantage.
