中医药干预PI3K/Akt信号通路防治心力衰竭作用机制的研究进展

摘要 心力衰竭是多种心血管疾病的最终结局,具有极高的致死率。磷脂酰肌醇-3-激酶(PI3K)/蛋白激酶B(PKB/Akt)信号通路是心力衰竭发病机制的重要调节途径,在改善血管舒张功能,抗炎、抗氧化应激,减轻心肌损伤、肥大、纤维化及心肌细胞凋亡,逆转心肌重构中发挥重要作用。中医药在防治心力衰竭方面具有多途径、多靶点及整体调控的优势,可通过干预PI3K/Akt信号通路抑制心肌纤维化、减轻心肌肥厚、抗心肌细胞凋亡、抗炎、抗氧化应激、减少心肌损伤及改善心肌重构。深入研究中医药干预PI3K/Akt信号通路防治心力衰竭的作用机制,可为中医药防治心力衰竭的临床研究及新药方的开发利用提供新的理论依据与辨证思路。 Heart failure is the final outcome of a variety of cardiovascular diseases,with a very high mortality.The phosphatidylinositol-3-kinase(PI3K)/protein kinase B(PKB/Akt)signaling pathway is an important regulatory pathway in the pathogenesis of heart failure,which plays an important role in improving vasodilation function,anti-inflammation,anti-oxidative stress,reducing myocardial injury,hypertrophy,fibrosis and cardiomyocyte apoptosis,and reversing myocardial remodeling.Traditional Chinese medicine has the advantages of multi-channel,multi-target and global regulation in the prevention and treatment of heart failure,which can inhibit myocardial fibrosis,reduce myocardial hypertrophy,resist myocardial cell apoptosis,inflammation,oxidative stress,reduce myocardial injury and improve myocardial remodeling by interfering with the PI3K/Akt signaling pathway.An in-depth study on the mechanism of action of traditional Chinese medicine intervention in PI3K/Akt signaling pathway for the prevention and treatment of heart failure,can provide new theoretical basis and dialectical thinking for the clinical research and the development and utilization of new prescriptions.
