新疆5种臭阿魏对小鼠宫颈癌U14细胞的体内外抑制作用研究 被引量:1

摘要 目的研究新疆5种臭阿魏乙醇提取物的体内及体外对鼠源性宫颈癌U14细胞的抑制作用。方法制备新疆5种臭阿魏根的乙醇提取物,MTT比色法检测5种臭阿魏乙醇提取物的不同浓度组(9.375、18.75、37.5、75、150、300μg/mL)对小鼠宫颈癌U14细胞的增殖抑制作用。建立宫颈癌U14小鼠荷瘤模型,随机分组后连续灌胃给药12 d,剥取肿瘤组织,计算各组抑瘤率。检测各组小鼠血清中丙氨酸氨基转移酶(ALT)、血肌酐(Cre)、胱抑素C(Cyc C)的值。HE染色观察各组肿瘤病理组织形态。结果大果阿魏、新疆阿魏、托里阿魏、阜康阿魏、圆锥茎阿魏乙醇提取物对小鼠宫颈癌U14细胞的IC_(50)分别为50.84、59.55、133.4、181.7、202.2μg/mL,提示大果阿魏抑制作用最强,新疆阿魏次之,托里阿魏、阜康阿魏、圆锥茎阿魏的抑制作用较差。大果阿魏与新疆阿魏高、低剂量组小鼠的瘤重与模型组相比较明显减轻(P<0.05)。5种臭阿魏均有一定抑瘤作用,其中大果阿魏高、低剂量组及新疆阿魏高、低剂量组的抑瘤效果更好,抑瘤率分别为47.12%、45.19%、49.10%、40.13%。与正常对照组相比,各阿魏给药组对小鼠肝、肾功能均无明显影响,顺铂组的胱抑素C显著升高,差异有统计学意义(P<0.05)。结论大果阿魏及新疆阿魏的乙醇提取物抗宫颈癌活性好。 Objective To study the inhibitory effects of ethanol extracts of 5 species of smelly Ferula from Xinjiang on mouse cervical cancer U14 cells in vivo and in vitro.Methods The ethanol extracts of Fructus Ferula root from Xinjiang were prepared,and the inhibitory effects of 5 kinds of ethanol extracts(9.375,18.75,37.5,75,150,300μg/mL)on the proliferation of mouse cervical cancer U14 cells were detected by MTT colorimetry.The tumor model of cervical cancer U14 mice was established and given by intragastric administration for 12 days after random grouping.The tumor tissue was stripped off and the tumor inhibition rate of each group was calculated.The serum levels of alanine aminotransferase(ALT),serum creatinine(Cre)and cystatin C(CycC)were measured.The histopathology of the tumor was observed by HE staining.Results The IC50 of ethanol extracts of F.lehmannii,F.sinkiangensisi,F.krylovii,F.fukangensisi and F.conocaula on mouse cervical cancer U14 cells were 50.84,59.55,133.4,181.7,202.2μg/mL,respectively,suggesting that the inhibitory effect of F.lehmannii was the strongest,followed by F.sinkiangensisi,and the inhibitory effects of F.krylovii,F.fukangensisi and F.conocaula were poor.The tumor weight of mice in the high and low dose groups of F.lehmannii and F.sinkiangensisi was significantly less than that in the model group(P<0.05).5 kinds of smelly Ferula all had certain anti-tumor effects,among which the high and low dose groups and the high and low dose groups of F.lehmannii and F.sinkiangensisi had better anti-tumor effect,and the tumor inhibition rates were 47.12%,45.19%,49.10%and 40.13%,respectively.Compared with the normal control group,each Ferula group had no significant effect on liver and renal function,but cystatin C in cisplatin group was significantly higher than that in normal control group(P<0.05).Conclusion The ethanol extracts of F.lehmannii and F.sinkiangensisi have better anti-cervical cancer activity.
