非洲猪瘟病毒抑制猪繁殖与呼吸综合征病毒体外复制的研究

Study on replication of porcine reproductive and respiratory syndrome virus is inhibited by African swine fever virus in vitro

摘要 非洲猪瘟(African swine fever,ASF)和猪繁殖与呼吸综合征(porcine reproductive and respiratory syndrome,PRRS)是由非洲猪瘟病毒(African swine fever virus,ASFV)和猪繁殖与呼吸综合征病毒(porcine reproductive and respiratory syndrome virus,PRRSV)引起的猪病毒性传染病,对全球养猪业造成严重危害。随着PRRSV与ASFV的流行,增加了二者混合感染的可能性。前期工作已表明PRRSV在体外可以抑制ASFV复制。然而,ASFV是否影响PRRSV体外复制尚不可知。因此,本研究通过实时荧光定量PCR(RT-qPCR)和免疫印迹试验(Western-blot)检测了PRRSV疫苗株JXA1-R和ASFV弱毒株混合感染后PRRSV在猪肺泡巨噬细胞(porcine alveolar macrophages,PAMs)和猪骨髓巨噬细胞(bone marrow-derived macrophages,BMDMs)的复制水平以及CD163的mRNA水平。结果显示,混合感染抑制了PRRSV的复制水平,并降低CD163的mRNA水平。本研究证明ASFV可抑制PRRSV在体外的复制,并抑制CD163的mRNA水平。该结果为PRRSV和ASFV混合感染的防控提供了理论依据。 African swine fever(ASF)and porcine reproductive and respiratory syndrome(PRRS)are viral diseases of pigs caused by African swine fever virus(ASFV)and porcine reproductive and respiratory syndrome virus(PRRSV),respectively.They pose a serious threat to the global swine industry.The coexistence and interaction of PRRSV and ASFV have increased the likelihood of co-infections.Previous work has shown that PRRSV can inhibit ASFV replication in vitro.However,it is unknown whether ASFV affects PRRSV replication in vitro.Therefore,this study investigated the mRNA level of CD163 of PRRSV vaccine strain JXA1-R and attenuated ASFV strain in co-infected porcine alveolar macrophages(PAMs)and bone marrow-derived macrophages(BMDMs)using real-time fluorescence quantitative PCR(RT-qPCR)and western blot analysis to detect the replication levels.The results showed that co-infection inhibited PRRSV replication and downregulated CD163 mRNA levels.This study demonstrates that ASFV can inhibit PRRSV replication and reduce CD163 mRNA levels in vitro,offering a theoretical basis for the prevention and control of co-infections of PRRSV and ASFV.
