切除修复交叉互补基因1血管内皮生长因子受体-3血小板衍生生长因子受体B在胃癌中的表达及意义

摘要 目的分析切除修复交叉互补基因1(ERCC1)、血管内皮生长因子受体-3(VEGFR3)、血小板衍生生长因子受体B(PDGFRB)在胃癌中的表达及意义。方法选取2019年6月至2021年6月河南科技大学第一附属医院病理科归档资料齐全完整的52例胃癌组织石蜡标本和距离癌组织边缘5cm的癌旁组织石蜡标本,采用免疫组织化学、实时荧光定量法检测ERCC1、VEGFR3、PDGFRB表达,分析ERCC1、VEGFR3、PDGFRB在胃癌中的表达变化及三者相关性。结果与癌旁组织相比,癌组织中ERCC1、VEGFR3、PDGFRB表达量升高,差异具有统计学意义(P<0.05)。ERCC1、VEGFR3、PDGFRB表达与胃癌患者的性别、年龄、肿瘤长径无关,与TNM分期、分化程度、淋巴结转移、浸润深度、病理类型相关,Ⅲ期、Ⅳ期、中低分化、有淋巴结转移、≥1/2肌层浸润、肠型胃癌患者ERCC1、VEGFR3、PDGFRB表达升高,差异具有统计学意义(P<0.05)。ERCC1、VEGFR3、PDGFRB之间Pearson相关性分析显示,ERCC1与VEGFR3之间正相关(r=0.572,P=0.001);ERCC1与PDGFRB之间正相关(r=0.617,P=0.001);VEGFR3与PDGFRB之间正相关(r=0.569,P=0.001)。ROC曲线显示,与ERCC1、VEGFR3、PDGFRB单项诊断相比,三项联合对胃癌的诊断价值较高(P<0.05)。结论ERCC1、VEGFR3、PDGFRB在胃癌组织中表达升高,并随着淋巴结的转移,ERCC1、VEGFR3、PDGFRB表达升高,参与疾病的发展。 Objective To analyse the expression and significance of excision-repair cross-complementation gene 1(ERCC1),vascularendothelial growth factor receptor-3(VEGFR3),platelet-derived growth factor receptor B(PDGFRB)in gastric cancer.Methods Paraffin specimens of 52 cases of gastric cancer tissues and paracarcinoma tissues 5 cm away from the edge of cancerous tissues with complete and complete data archived in the Department of Pathology of irst Affiliated Hospital of Henan Vniversity of Science and Technology,from June 2019 to June 2021 were selected.Immunohistochemistry and real-time fluorescence quantification were used to detect the expression of ERCC1,VEGFR3 and PDGFRB,and to analyse the changes in the expression of ERCC1,VEGFR3 and PDGFRB and the correlation between the three in gastric cancer.Results The expressions of ERCC1,VEGFR3 and PDGFRb in cancer tissues were significantly higher than those in adjacent tissues(P<0.05).The expression of ERCC1,VEGFR3 and PDGFRb was not related to gender,age and tumor diameter and of patients with gastric cancer,but associated to TNM stage,degree of differentiation,lymph node metastasis,depth of invasion and pathological type.The expression of ERCC1,VEGFR3 and PDGFRb in patients with stageⅢ,Ⅳ,medium and low differentiation,lymph node metastasis,≥1/2 myometrial invasion and intestinal gastric cancer was significantly higher(P<0.05).Pearson correlation analysis among ERCC1,VEGFR3 and PDGFRb showed that they have the same relationship in the same direction ERCC1 and VEGFR3(r=0.572,P=0.001);they have the same relationship in the same direction ERCC1 and PDGFRb(r=0.617,P=0.001);they have the same relationship in the same direction VEGFR3 and PDGFRb(r=0.569,P=0.001).ROC curve showed that the combination of ERCC1,VEGFR3,and PDGFRB had a higher diagnostic value for gastric cancer compared to ERCC1,VEGFR3,and PDGFRB alone(P<0.05).Conclusion The expression of ERCC1,VEGFR3 and PDGFRB is increased in gastric cancer tissues,and intensifies with lymph node metastasis,participating in disease progression.
