绿花白千层精油化学成分及宫颈炎活性研究

Chemical constituents of essential oil from Melaleuca viridiflora and related cervicitis activity

目的研究4种绿花白千层(Melaleuca viridiflora Sol.ex Gaertn)精油的化学成分及其治疗宫颈炎的活性。方法采用气相色谱-质谱联用仪(GC-MS)鉴定其化学成分,通过阴道灌注酚类黏液建立大鼠宫颈炎模型,研究绿花白千层精油对宫颈炎的治疗作用。给药后1周监测外阴炎症、脏器、宫颈炎血清环氧合酶2(COX-2)、超氧化物歧化酶(SOD)、丙二醛(MDA)、血浆铜蓝蛋白(CP)水平,确定精油的功效。结果对4种绿花白千层精油Yk-2、Yk-10、Yk-11、Yk-17进行了分析,发现分别含有34个、25个、28个和21个化合物,鉴定率均在90%以上。主要成分是桉油精、α-蒎烯、α-松油醇、β-蒎烯、γ-萜品烯、α-萜品醇、β-石竹烯和喇叭茶醇。研究发现,以高、中、低剂量给药的Yk-2精油栓剂均可降低模型大鼠的外阴炎症评分,与正常组相比,可改善宫颈炎指数和血清生化因子COX-2、SOD、MDA和CP水平。结论4种绿花白千层精油成分丰富,鉴别率高,Yk-2精油栓剂对大鼠宫颈和阴道组织炎症表现出良好的治疗作用。

Objective To determine the chemical constituents of essential oil from Melaleuca viridiflora and related cervicitis activity.Methods Gas chromatography-mass spectrometry(GCMS)was used to identify the chemical components,while rat cervicitis models were established through vaginal perfusion of phenol mucilage to determine the therapeutic effect of essential oil from Melaleuca viridiflora on cervicitis.The levels of serum cyclooxygenase-2(COX-2),superoxide dismutase reagent(SOD),malondialdehyde reagent(MDA),and ceruloplasmin reagent(CP)in vulva inflammation,organs,and cervicitis were monitored one week after the administration to determine the efficacy of essential oil.Results Four types of Melaleuca viridiflora essential oils Yk-2,Yk-10,Yk-11,and Yk-17 were analyzed and found 34,25,28,and 21 chemical constituents were with identification rates above 90%.The main constituents included eucalyptol,α-pinene,β-pinene,γ-terpinene,α-terpineol,β-caryophyllene,and trumpet alcohol.The Yk-2 essential oil suppository,administered at high,medium,and low dosages,reduced vulvar inflammation scores in the model rats and improved cervicitis index and the serum levels of COX-2,SOD,MDA and CP,as compared with those of the normal group.Conclusion The four types of Melaleuca viridiflora essential oils have rich components and high identification rates.The Yk-2 essential oil suppository shows good therapeutic effect on rat cervical and vaginal tissue inflammation.
