摘要
目的采用超高效液相色谱-四极杆-静电场轨道阱(UPLC-Q-Exactive Orbitrap-MS),对清热祛湿凉茶的化学成分进行快速识别和鉴定。方法采用Luna C18色谱柱(100 mm×2.1 mm,1.8μm),0.1%甲酸水溶液-乙腈溶液为流动相,梯度洗脱,流速为0.3 mL·min^(-1),柱温为35℃,采用正负离子检测模式。结果共鉴定出清热祛湿凉茶中33种化学成分。包括糖类成分2个,有机酸类成分5个,香豆素类成分1个,黄酮类成分16个,苯丙素类成分4个和三萜类成分5个。结论本研究首次采用UPLC-Q-Exactive Orbitrap-MS技术对清热祛湿凉茶的主要成分进行鉴定,可为清热祛湿凉茶的药效成分研究提供科学依据。
Objective To identify the chemical constituents in Qingre Qushi herbal tea by UPLCQ-Exactive Orbitrap-MS.Methods Luna C18 Column(100 mm×2.1 mm,1.8μm)was used with 0.1%formic acid solution and acetonitrile as the mobile with gradient elution.The flow rate was 0.3 mL·min^(-1)and the column temperature was 35℃.The mass spectrometry adopted the positive and negative mode.Results Totally 33 chemical constituents in Qingre Qushi herbal tea were identified,including 2 sugars,5 organic acids,1 coumarin,16 flavonoids,4 phenylpropanoids and 5 triterpenoids.Conclusion UPLC-Q-Exactive Orbitrap-MS technique has been established for the first time to identify the main components in Qingre Qushi herbal tea,providing a scientific basis for the study of pharmacodynamic components in Qingre Qushi herbal tea.
