不同居群北柴胡ISSR遗传多样性分析 被引量:1

摘要 利用简单重复间序列(inter-simple sequence repeat,ISSR)分子标记技术对10个不同居群的北柴胡样品进行遗传多样性分析,为进一步开展北柴胡优良种质的选育提供依据。以不同居群的北柴胡新鲜叶片为材料,用植物组织DNA提取试剂盒提取北柴胡样品DNA。利用单因素梯度实验研究最佳ISSR-PCR反应体系,将不同居群的DNA扩增产物进行琼脂糖凝胶电泳,利用NTSYS-pc软件分析不同居群北柴胡的遗传多样性。结果发现,10个居群的北柴胡遗传相似系数介于0.1428~1.0000。经聚类分析,10个不同居群的北柴胡分为3大类,第1类为甘肃北柴胡;第2类包括5个北柴胡样品,分别为陵川、河南、闻喜、平陆、襄汾北柴胡,均属于山西省晋南居群;第3类包括4个北柴胡样品,分别为内蒙古、临汾、浑源、广灵北柴胡,类属于山西省北部居群及内蒙古居群;10个居群的北柴胡存在比较丰富的遗传多样性,且与其地理环境有一定的关联性。研究结果可为北柴胡种质资源保护及优良种质选育提供理论依据。 The genetic diversity of 10 different populations of Bupleurum chinense was studied by inter-simple sequence repeat(ISSR)molecular marker technique,in order to provide basis for further breeding of B.chinense.The DNA of Bupleurum chi⁃nense samples from fresh leaves of different populations was extracted by the plant tissue DNA extraction kit.The single factor gradient experiment was used to study the optimal ISSR-PCR reaction system.The DNA amplification products of different populations were subjected to agarose gel electrophoresis,and the genetic diversity of different populations of B.chinense was analyzed by NTSYS-pc software.The results showed that the genetic similarity coefficient of 10 populations ranged from 0.1428 to 1.0000.10 different populations were divided into three categories by cluster analysis,with the first category being Gansu.The second category included five samples of B.chinense,Lingchuan,Henan,Wenxi,Pinglu,and Xiangfen,all belonging to the Jinnan population in Shanxi Province.The third category included four samples,namely Inner Mongolia,Linfen,Hunyuan,and Guangling,belonging to the northern population of Shanxi Province and Inner Mongolia.The 10 populations of B.chinense have relatively abundant genetic diversity and are related to their geographical environment.The results could provide theoretical basis for germplasm resources protection and elite germplasm breeding of B.chinense.
