外源植物生长调节剂对桑黄发酵产黄酮的影响

摘要 黄酮是药用真菌桑黄最为重要的活性成分之一,为探究植物生长调节剂对桑黄液体发酵培养中黄酮产量的影响,以6-苄氨基腺嘌呤(6-BA)、激动素(KT)、吲哚丁酸(IBA)、水杨酸(SA)、2,4-二氯-苯氧乙酸(2,4-D)、萘乙酸(NAA)、赤霉素(GA_3)和脱落酸(ABA)等8种植物生长调节剂作为外源添加物,测定不同处理桑黄液体发酵7 d、9 d、11 d的菌丝体生物量及胞内黄酮含量,并计算发酵11天时胞内黄酮总产量。结果表明,相比空白对照,6-BA、KT、IBA、SA、2,4-D能显著促进桑黄菌丝体生物量增加(P<0.05),但作用效果与发酵时间段有关,NAA无明显影响,GA_3和ABA引起桑黄菌丝体生物量显著下降;KT、6-BA、IBA和SA能显著提高桑黄胞内黄酮产量,NAA和2,4-D无明显作用,而GA_3和ABA则抑制桑黄黄酮代谢。综合比较,8种植物生长调节剂中以KT对桑黄菌丝体生物量和黄酮代谢的促进作用最明显,在桑黄液体发酵培养时添加KT有助提高黄酮产量。 In this study,the mycelial biomass,intracellular flavonoid content and total flavonoid yield of Sanghuangporus spp.were determined after liquid fermentation with 8 kinds of plant growth regulators as exogenous additives for 7 d、9 d and 11 d.The effects of plant growth regulators on the fermentation culture of Sanghuangporus spp.were investigated.The results showed that compared with CK,6-benzylamino-adenine(6-BA),kinetin(KT),indole butyric acid(IBA),salicylic acid(SA)and 2,4-dichloro-phenoxyacetic acid(2,4-D)could promote the mycelial biomass increase,but the effect was related to the fermentation time.Compared with CK,KT,6-BA,IBA and SA of the 8 plant growth regulators could significantly promote the yield of Sanghuangporus spp.flavone(P<0.05),while naphthalene acetic acid(NAA)and 2,4-D had no significant effect.However,gibberellin(GA3)and abscisic acid(ABA)inhibited the growth and flavonoid metabolism of Sanghuangporus spp..The stimulative effect of KT was the most obvious,and adding KT in liquid fermentation culture of Sanghuangporus spp.was helpful to increase the yield of flavonoids.
