摘要
目的:采用星点设计-响应面法优化盐覆盆子有效成分鞣花酸的提取工艺。方法:建立HPLC测定鞣花酸含量,流动相为乙腈-0.2%磷酸溶液(15:85);检测波长为254 nm,流速为1.0 ml/min,柱温为30℃;采用星点设计-响应面法对甲醇浓度、提取时间、液料比3个因素进行考察,优选盐覆盆子有效成分鞣花酸的提取工艺。结果:优选所得的盐覆盆子中鞣花酸提取工艺为甲醇浓度73%、提取时间63 min、液料比60∶1,此条件下鞣花酸含量为0.353%。结论:本试验优选所得的提取工艺高效稳定、操作简易、可用于盐覆盆子有效成分鞣花酸的提取。
Objective To optimize the extraction process of ellagic acid from Raspberry salina by central composite design response surface methodology.Methods HPLC was established to determine the content of ellagic acid.The mobile phase was acetonitrile-0.2%phosphoric acid solution(15∶85);The detection wavelength is 254 nm,the flow rate is 1.0 mL·min-1,and the column temperature is 30℃;Three factors,methanol concentration,extraction time and liquid to material ratio,were investigated by central composite design response surface methodology to optimize the extraction process of ellagic acid,an effective component of salt raspberry.Results The optimum extraction process of ellagic acid from salt raspberry is that the concentration of methanol is 73%,the extraction time is 63 min,and the ratio of liquid to material is 60∶1.Under this condition,the content of ellagic acid is 0.353%.Conclusion The optimized extraction process is efficient,stable and easy to operate.It can be used for the extraction of ellagic acid,an effective component of raspberry salt.
