HPLC法测定抗骨髓炎片中绿原酸和秦皮乙素的含量

Determination of Chlorogenic Acid and Esculetin in Kanggusuiyan Tablets by HPLC

目的:建立高效液相色谱(HPLC)法测定抗骨髓炎片中绿原酸、秦皮乙素含量的方法。方法:采用Thermo Hypersil Gold-C_(18)(4.6 mm×250 mm, 5μm)色谱柱,流动相为乙腈-0.1%磷酸溶液(9∶91),流速1.0 mL·min^(-1),柱温30℃,检测波长为327 nm,进样量为10μL。结果:绿原酸、秦皮乙素分别在14.46~144.6μg·mL^(-1)(r=0.9999)、3.137~31.37μg·mL^(-1)(r=0.9999)范围内线性关系良好,平均回收率分别为98.65%(RSD=0.75%)、96.51%(RSD=1.58%)。结论:所建立的方法准确,重现性好,可用于抗骨髓炎片中绿原酸和秦皮乙素含量的测定。

Objective To establish a method of determining chlorogenic acid and esculetin in Kanggusuiyan tablets by HPLC.Methods The samples was separated on the column Thermo Hypersil Gold-C 18(4.6 mm×250 mm,5μm),which the mobile phase was consisted of acetonitrile and 0.1%phosphoric acid at 9∶91.The flow rate was 1.0 mL·min^(-1).The column temperature was set at 30℃,the detection wavelength was 327 nm,and the injection volume was 10μL.Results Good linear relationships were fund for chlorogenic acid and esculetin in the range of 14.46-144.6μg·mL^(-1)(r=0.9999),3.137-31.37μg·mL^(-1)(r=0.9999),respectively.The average recoveries were 98.65%(RSD=0.75%)for chlorogenic acid and 96.51%(RSD=1.58%)for esculetin.Conclusion The method is accurate and reproducible,and can be applied in the content determination of chlorogenic acid and esculetin in Kanggusuiyan tablets.
