马铃薯StKNOX1基因的克隆及表达分析

摘要 KNOX基因是一类转录调控因子,其和植物的生长发育密切相关。为了探讨同源异型盒(KNOX)基因在马铃薯(Solanum tuberosum L.)块茎发育中的作用,采用RT-PCR和RACE技术,克隆了1个KNOX同源基因,且根据属性情况命名为StKNOX1,其属于KNOX家族I类蛋白,编码含344个氨基酸的蛋白,包含KNOX1、KNOX2、ELK等结构域。根据相关生物信息研究结果,该基因编码的蛋白与番茄中KNOX1的一致性最高(93.7%)。StKNOX1基因在马铃薯的各组织器官中均有表达,在根和块茎中的表达水平显著高于其他部位的表达水平。本研究将为进一步开展基因功能研究奠定理论基础,为马铃薯生物技术育种提供基因资源。 KNOX gene is a kind of transcriptional regulator,which is closely related to plant growth and development.In order to investigate the role of KNOX gene in tuber development of potato(Solanum tuberosum L.),a KNOX homolog gene was cloned by RT-PCR and RACE technology and named StKNOX1 according to its attributes.It belongs to the classⅠprotein of KNOX family.The gene encodes a protein containing 344 amino acids,including KNOX1,KNOX2,ELK and other domains.According to the results of related biological information study,the protein encoded by this gene had the highest consistency with KNOX1 in tomato(93.7%).The StKNOX1 gene was expressed in all tissues and organs of potato,and the expression level in roots and tubers was significantly higher than that in other parts.This study will lay a theoretical foundation for further research on gene function and provide genetic resources for potato biotechnology breeding.
