HSP70和S100A9蛋白在脑胶质瘤组织中的表达及其意义

摘要 目的 探讨热休克蛋白70(HSP70)和S100钙离子结合蛋白A9(S100A9)在胶质瘤组织中的表达水平及在低级别脑胶质瘤与胚胎发育不良性神经上皮肿瘤(DNT)中的鉴别诊断价值。方法 所有病理标本取自2015年1月至2020年12月广东祈福医院和南部战区总医院确诊的160例胶质瘤初诊患者,其中世界卫生组织(WHO)Ⅰ、Ⅱ、Ⅲ、Ⅳ级胶质瘤患者均为40例。采用免疫组织化学法检测160例胶质瘤和肿瘤旁正常脑组织,以及20例DNT组织中HSP70和S100A9的表达水平。结果 HSP70和S100A9在160例胶质瘤组织中的阳性率分别为85.0%(136/160)和7.5%(12/160)。HSP70在不同WHO分级胶质瘤组织中的表达水平比较,差异有统计学意义(P<0.05),且表达水平随着胶质瘤WHO级别的增加而升高,也能用于低级别胶质瘤与DNT的鉴别诊断。S100A9在不同WHO分级胶质瘤的表达水平比较,差异无统计学意义(P>0.05),也不能用于DNT与低级别胶质瘤的鉴别诊断。HSP70与S100A9在脑胶质瘤组织中的表达无相关性。结论 HSP70可用于脑胶质瘤的诊断,表达水平随着胶质瘤WHO级别的增加而升高,且可用于DNT与低级别胶质瘤的鉴别诊断。S100A9无上述价值。 Objective To research the expression level of heat shock protein 70(HSP70)and S100 calcium-binding protein A9(S100A9)in glioma tissue and their differential diagnostic value in low-level brain glioma and dysembryoplastic neuroepithelial tumor(DNT).Methods All pathological specimens were taken from 160 newly diagnosed patients with gliomas in Guangdong Clifford Hospital and Chinese People′s Liberation Army Southern Theater Command Gneral Hospital from January 2015 to December 2020,including 40 patients with World Health Organization(WHO)gradeⅠ,Ⅱ,ⅢandⅣgliomas.Immunohistochemical method was used to detect the expression levels of HSP70 and S100A9 in 160 cases of glioma and normal brain tissue adjacent to tumor,and 20 cases of DNT.Results The detectable rates of HSP70 and S100A9 were 85.0%(136/160)and 7.5%(12/160),respectively.The expression of HSP70 in gliomas with differences WHO grades was statistically significant(P<0.05),and the expression level increased with the improvement of who grade of gliomas,which could also be used for the differential diagnosis of low-grade gliomas and DNT.S100A9 had no statistical difference in different WHO grades(P>0.05),and it could not be used for the differential diagnosis of DNT and low-grade glioma.There was no correlation between HSP70 and S100A9 expression in glioma tissues.Conclusion HSP70 can be used in the diagnosis of brain glioma.The expression level increases with the improvement of WHO grade of glioma,and can be used in the differential diagnosis of low-level glioma and DNT.S100A9 has no such value.
