基于质谱技术对大豆低聚肽的多肽鉴别与质量研究

Peptide ldentification and Quality Research of Soy Oligopeptides Basic on the Mass-Spectrographic Technique

摘要 为鉴别大豆低聚肽中的多肽组分,并筛选适用于质量监控的多肽,通过四级杆串联飞行时间质谱法测定4批次大豆低聚肽样品。借助Proteo Wizard 3.0、Proteome Discoverer 2.4、Pep OS 1.0等多肽组学分析软件进行肽组学分析,鉴别了丰度前30的多肽,初筛出21种适宜于大豆低聚肽质量监控的短肽;优选具有全匹配的8种短肽,利用液相色谱-三重四极杆质谱联用技术对8种短肽进行方法学研究。结果表明8种低聚肽中仅VV和VN适宜合于建立大豆低聚肽的质量监控。VV和VN的线性相关系数R值均大于0.99、回收率高、精密度好。建立的VV和VN检测方法稳定可靠,VV和VN在大豆低聚肽中稳定存在,可作为大豆低聚肽原料及相关产品中特征性短肽,用于产品质量控制的目标。 In order to identify the polypeptide compositions of soy oligopeptides,and to filtrate the suitable polypeptide for quality monitoring.The study used the Quadrupole-Time of Flight(Q-TOF)to measure 4 batches of soy oligopeptide samples.This research used the analysis software of peptidomics(include:Proteo Wizard 3.0,Proteome Discoverer 2.4,Pepos 1.0 et.al.)to identify the polypeptide of 30 highest abundance,and primary screening 21 short peptides that suitable to monitor the quality of soy oligopeptides.And 8 short peptides were chosen to become the target of quality control that possessing identification of an exact match,the mass-spectrographic technique of liquid chromatography-mass spectrometry-mass spectrometer(LC-MS-MS)was used to conduct methodological study with 8 types of short peptides.The result show only 2 peptides(VV&VN)were suitable use to build up quality monitoring of soy oligopeptides.The linear correlation R value for VV&VN was higher than O.99,and their recovery rate and precision were high.In addition,the detection method for VV&VN is reliable and stable.Both of VV&VN were stable in soy oligopeptides.They can become the target of quality monitoring for distinctive peptides in the raw material of soy oligopeptides and other related products.
