摘要
为研究氟对斑马鱼胚胎/仔鱼发育的毒性效应,将斑马鱼胚胎暴露于0、20、40和80 mg·L^(-1)的氟化钠溶液中,观察并记录孵化率、畸形率、死亡率、心率,测定其超氧化物歧化酶(superoxide dismutase,SOD)、过氧化氢酶(Catalase,CAT)、丙二醛(Malonic dialdehyde,MDA)、活性氧(reactive oxygen species,ROS)等抗氧化参数,用酶联免疫吸附法检测了其T3和T4的含量,并用实时荧光定量PCR方法检测HPT轴上内分泌相关基因的表达.结果显示,氟化钠可导致斑马鱼死亡率和畸形率上升而孵化率和心率下降,T3和T4含量、抗氧化参数和HTP轴相关基因表达的显著变化.综上,氟化钠可通过影响斑马鱼甲状腺激素水平、抗氧化参数及HPT轴上内分泌相关基因的表达对其胚胎/仔鱼发育产生毒性效应.本实验结果对进一步探究水环境中氟的生态毒性效应及风险性评价提供了理论支持.
To assess the detrimental impact of fluoride on the development of zebrafish embryos/larvae,embryos were exposed to varing concentrations of fluoride(0,20,40,80 mg·L^(-1))for 9 d to observe the hatching rate,malformation rate,mortality rate,heart rate,determine the antioxidant parameters including superoxide dismutase(SOD),catalase(CAT),malonic dialdehyde(MDA),reactive oxygen species(ROS),the levels of T3 and T3 using enzyme-linked immunosorbent assay,and the expression of endocrine related genes in the HPT axis using real-time fluorescence quantitative PCR.The results showed that fluoride could increase the mortality rate and malformation rate of zebrafish,while simultaneously decrease the hatching rate and heart rate.Moreover,fluoride exposure resulted in significant changes in the levels of thyroid hormones(T3 and T4),antioxidant indexes,and the gene expression in the HPT axis.In conclusion,this study highlights the toxic effects of fluoride on zebrafish embryos/larva development by disturbing the levels of thyroid hormones,antioxidant parameters,and the expression of endocrine related genes in the HPT axis.The results can provide essential theoretical support for further investigation into the ecological toxicity effects and risk assessment of fluoride in the water environment.
