薏苡仁多糖的分离纯化及抗氧化活性研究 被引量:3

摘要 以脱脂薏苡仁为原料,采用水提醇沉法制备薏苡仁多糖,通过DEAE Sepharose Fast Flow离子交换柱层析,得到薏苡仁多糖CSP1和CSP2 2种组分,结合扫描电镜和红外光谱对其结构进行分析,并通过对氧化应激细胞的保护作用研究其抗氧化活性。结果表明:纯化后的薏苡仁多糖CSP1和CSP2总糖含量分别为90.83%和88.66%,蛋白质含量分别为1.403%和0.086%。体外抗氧化试验显示薏苡仁多糖CSP1和CSP2能提高H_(2)O_(2)诱导的WRL-68人正常肝细胞内超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和谷胱甘肽过氧化物酶(GSH-Px)的活力,起到抗氧化应激保护的作用,为薏苡仁多糖的应用提供理论依据。 Using defatted coix seed as raw material,coix seed polysaccharide was prepared by water ex-traction and alcohol precipitation method.Two components of coix seed polysaccharide CSP1 and CSP2 were obtained by DEAE Sepharose Fast Flow ion exchange chromatography.Their structures were ana-lyzed by scanning electron microscopy and infrared spectroscopy,and their antioxidant activities were studied through the protective effect on oxidative stress cells.The results showed that the total sugar con-tents of coix seed polysaccharide CSP1 and CSP2 were 90.83%and 88.66%,respectively,and the protein contents were 1.403%and 0.086%,respectively.In vitro antioxidant test showed that coix seed polysaccharides CSP1 and CSP2 could in prove the actirities of superoxide dismutase(SOD),cata-lase(CAT)and glutathione peroxidase(GSH-Px)activities in WRL-68 human normal hepatocytes in-duced by H_(2)O_(2),and play a protective role against stress,providing theoretical basis for the application of coix seed polysaccharides.
