陕西产凤尾七HPLC指纹图谱研究及4种成分定量分析

摘要 目的建立凤尾七的高效液相色谱(HPLC)指纹图谱与含量测定方法。方法采用Ultimate LP⁃C18(4.6 mm×250 mm,5μm)色谱柱,以甲醇和体积分数0.1%磷酸水溶液为流动相,梯度洗脱,检测波长为274 nm,建立10批陕西产凤尾七的指纹图谱。利用主成分分析(PCA)进一步对影响凤尾七药材质量均一性的差异性成分进行筛选。基于指纹图谱及差异成分,选取凤尾七质量控制指标成分,进行含量测定。结果10批凤尾七指纹图谱相似度均大于0.993,确定共有峰15个,其中4个共有峰被指认。PCA筛选出2个主成分,方差累积贡献率为84.3%;影响凤尾七批间质量均一性的主要组分有没食子酸、芦丁等。测得10批凤尾七中没食子酸、芦丁、草质素苷和山柰酚⁃7⁃O⁃鼠李糖苷的含量分别为0.2159~0.3901、0.2132~0.3445、3.2214~4.9397和0.3069~0.4458 mg·g^(-1)。结论建立的指纹图谱与含量测定方法稳定、可控,适用于陕西产凤尾七药材的质量控制。 OBJECTIVE To establish the HPLC fingerprint and content determination method for Rhodiola dumulosa.METH⁃ODS The fingerprints of 10 batches of Rhodiola dumulosa from Shaanxi province were established using Ultimate LP⁃C18(4.6 mm×250 mm,5μm)column with gradient elution of methanol and 0.1%phosphoric acid aqueous solution as mobile phases,and the de⁃tection was conducted at 274 nm.Principal component analysis(PCA)was used to further screen the differential components that af⁃fected the quality uniformity of Rhodiola dumulosa.Based on the fingerprint and differential components,the quality control index com⁃ponents of Rhodiola dumulosa were selected for content determination.RESULTS The similarity of the fingerprints based on 10 bat⁃ches of Rhodiola dumulosa was all more than 0.993.A total of 15 common peaks were calibrated,of which four common peaks were i⁃dentified.Two principal components were screened by PCA,and the cumulative contribution rate of variance was 84.3%.Gallic acid,rutin and other unidentified components were the main components that affected the quality uniformity between batches.The contents of gallic acid,rutin,rhodionin and kaempfer⁃7⁃O⁃rhamnoside in 10 batches of Rhodiola dumulosa were 0.2159-0.3901,0.2132-0.3445,3.2214-4.9397 and 0.3069-0.4458 mg·g^(-1),respectively.CONCLUSION The established fingerprint and con⁃tent determination method are stable and controllable,and can be used for quality control of Rhodiola dumulosa from Shaanxi province.
