2022年山东地区商品肉鸡15株H9N2亚型禽流感病毒HA基因的遗传演化分析

摘要 为了解2022年商品肉鸡H9N2亚型禽流感病毒在山东地区的流行情况,选取了15株分离鉴定的H9N2亚型禽流感毒株,对其HA基因进行序列测定和分子特征分析,并应用软件Mega6.0绘制HA基因进化树,进行遗传进化分析。结果表明,15株H9N2分离毒的HA裂解位点氨基酸均为RSSR/GLF,具有典型低致病性AIV HA基因的特征;其第234位氨基酸均为亮氨酸(L),因此就具有了结合人流感受体的分子特性。进化树分析表明,2022年山东地区15株H9N2流行毒株HA基因属于4.2.5分支,细分为近几年在中国鸡群中流行的4.2.5a和4.2.5.c亚分支。 In order to understand the epidemic situation of commercial broilers H9N2 subtype avian influenza virus in Shandong region in 2022,15 strains of H9N2 subtype avian influenza virus were selected,sequenced and molecularly analysed for its HA gene,and applied the software Mega6.0 to draw the HA gene evolution tree to analyse genetic evolution.The results show that the amino acids at the HA cleavage site of 15 strains of H9N2 both are RSSR/GLF,which have the characteristics of a typical low pathogenic AIV HA gene;its 234th amino acid is leucine(L),consequently it has the molecular characteristics of binding human flow receptors.Evolutionary tree analysis shows that the HA gene of 15 H9N2 epidemic strains in Shandong in 2022 belongs to the 4.2.5 branch,which is sub-branched into 4.2.5a and 4.2.5.c,which are popular in Chinese chicken flock in recent years.
