摘要
目的确定在乙型肝炎病毒表面抗原(HBsAg)定量检测中影响稀释结果的主要物质成分,为HBsAg稀释液的选择提供思路。方法分别使用生理盐水,50 g/L小牛血清白蛋白(BSA)、15 g/L小牛血清球蛋白(BGG)以及阴性血清对101份HBsAg阳性样本进行3倍稀释,计算稀释带来的偏差。分别将BSA和BGG配制成不同浓度稀释液,对10份HBsAg阳性样本进行3倍稀释,分析稀释后检测结果差异。分别使用生理盐水,50 g/L的BSA、15 g/L的BGG以及阴性血清对15例HBsAg阳性样本进行3倍稀释,然后分别将反应总时间控制在32 min、62 min、77 min和92 min,分别计算62 min、77 min和92 min检测结果相对于32 min检测结果的比值(即增幅),比较不同稀释组在92 min时的增幅。结果15 g/L的BGG稀释所得的回收率偏差最小。BSA不同浓度之间的稀释结果无明显差异,而BGG不同浓度之间稀释结果成负相关。15 g/L的BGG稀释组在92 min相对于32 min的检测浓度的增幅最大。结论15 g/L BGG作为HBsAg稀释液效果较为理想。
Objective Determine the main substance components that affect the dilution results in the quantitative detection of hepatitis B virus surface antigen(HBsAg),and provide ideas for the selection of HBsAg dilutions.Methods A total of 101 HBsAg positive samples were diluted three times with normal saline,50 g/L bovine serum albumin(BSA),15 g/L bovine serum globulin(BGG)and negative serum respectively,and the deviation caused by dilution was calculated.BSA and BGG were prepared into different concentrations of diluents,and 10 HBsAg positive samples were diluted three times,and the difference of detection results after dilution was analyzed.Fifteen HBsAg positive samples were diluted three times with normal saline,50 g/L bovine serum albumin(BSA),15 g/L bovine serum globulin(BGG)and negative serum,respectively.Then the total reaction time was controlled at 32 min,62 min,77 min and 92 min,respectively.The ratio of the results of 62 min,77 min and 92 min to the results of 32 min(i.e.,increase)was calculated,and the increase of different dilution groups at 92 min was compared.Results The deviation of 15 g/L BGG group was the smallest.There was no significant statistical difference compared with the negative serum dilution group,while the dilution results between different concentrations of BGG are negatively correlated.The 15 g/L BGG group has the largest increase in the detection concentration at 92 min compared to 32 min.Conclusion 15 g/L BGG is more effective as HBsAg diluent.
