大黄炭炮制过程中“增效”和“减毒”潜在质量标志物的变化规律研究

摘要 目的:通过筛选大黄炭的潜在质量标志物,分析大黄炭炮制过程中潜在质量标志物的变化规律,为大黄炭炮制机理研究与炮制终点判断提供参考。方法:本研究基于经典文献与网络药理学分析,初步筛选大黄炭的潜在质量标志物。以LPS诱导的HUVEC细胞作为大黄炭“凉血化瘀”功效的体外评价模型,进一步验证大黄炭的潜在质量标志物。使用色差仪对大黄炭的颜色特征进行量化,分析大黄炭炮制过程中潜在质量标志物与颜色变化的关联性。结果:大黄炭的潜在质量标志物可分为两类:结合蒽醌与番泻苷为一类,随炮制程度增加而递减,可能与大黄制炭后泻下作用减弱有关;游离蒽醌、没食子酸、5-HMF为另一类,随炮制程度增加而先增后减,可能与大黄制炭后凉血化瘀作用增加有关。结论:大黄炭中结合蒽醌可作为潜在的“减毒”质量标志物,游离蒽醌可作为潜在的“增效”质量标志物。 Objective:To screen potential quality markers of rhubarb charcoal and analyze the changes in potential quality markers during the processing of rhubarb charcoal,providing reference for the study of the processing mechanism of rhubarb charcoal and the determination of the processing endpoint.Methods:Based on network pharmacology and literature mining,the potential quality markers of rhubarb charcoal were preliminarily screened.The LPS induced vascular endothelial cells(HUVEC)were used as the model of TCM efficacy of"cooling blood and removing blood stasis".The colorimeter was used to study the colority of samples heated under different conditions,and to analyze the correlation between potential quality markers processing of rhubarb charcoal and color values.Results:The potential quality markers of rhubarb charcoal were divided into two categories.The first is the one-way decrease of potential quality markers with purgative effect,including the combination of anthraquinone and sennoside,and the second is the first increase and then decrease of potential quality markers for anti-inflammatory and hemostasis,including free anthraquinone,gallic acid and 5-HMF.Conclusion:The combined anthraquinone in rhubarb charcoal can be used as the quality marker of"detoxification",and the free anthraquinone can be used as the quality marker of"enhancement".
