摘要
目的探究人脐带间充质干细胞(hUC-MSCs)来源的神经干细胞样细胞(NSCLCs)对大脑中动脉闭塞/再灌注(MCAO/R)损伤的保护作用。方法将hUC-MSCs诱导成NSCLCs。选取250~280 g雄性Sprague Dawley大鼠30只,随机分为假手术组(Sham组)、模型组(MCAO/R组)和治疗组(NSCLCs组)。采用线栓法构建MCAO/R损伤模型,NSCLCs组造模后采用经颈外-颈内动脉注射移植NSCLCs治疗。通过改良神经功能缺损评分(mNSS评分)评估各组大鼠的神经行为功能,苏木精-伊红(HE)染色观察神经元形态结构,免疫组化染色观察神经元核蛋白(NeuN)表达。结果MCAO/R组大鼠mNSS评分较Sham组显著下降,而NSCLCs组较MCAO/R组显著升高(F=5.119,P<0.05);MCAO/R组损伤神经细胞较Sham组增多,而NSCLCs组较MCAO/R组显著减少(F=173.928,P<0.01);NeuN阳性细胞数目MCAO/R组较Sham组显著降低,而NSCLCs组较MCAO/R组显著升高(F=73.120,P<0.01)。结论NSCLCs移植可通过改善神经元细胞损伤对大鼠脑缺血再灌注损伤发挥保护作用。
Objective To investigate the protective effect of neural stem cell-like cells(NSCLCs)derived from human umbilical cord mesenchymal stem cells(hUC-MSCs)againstmiddle cerebral artery occlusion/reperfusion(MCAO/R)injury in rats.Methods The hUC-MSCs were induced into NSCLCs.A total of 30 male Sprague-Dawley rats weighing 250-280 g were randomly divided into sham-operation group(Sham group),model group(MCAO/R group),and treatment group(NSCLCs group).The suture method was used to establish a rat model of MCAO/R,and after modeling,the rats in the treatment group weretreated with NSCLCs transplantation via the external-internal carotid artery.The modified Neurological Severity Score(mNSS)was used to evaluate theneurobehavioral function of rats in each group,HE staining was used to observe neuronal morphology and structure,and immunohistochemical staining was used to measure the expression of neuronal Neurexin(NeuN).Results The MCAO/R group had a significant reduction in mNSS score compared with the Sham group,while the NSCLCs group had a significant increase compared with the MCAO/R group(F=5.119,P<0.05).Compared with the Sham group,the MCAO/R group had a significant increase in the number of injured neuronal cells,and compared with the MCAO/R group,the NSCLCs group had a significant reduction in the number of such cells(F=173.928,P<0.01).Compared with the Sham group,the MCAO/R group had a significant reduction in the number of NeuN-positive cells,and compared with the MCAO/R group,the NSCLCs group had a significant increase in the number of such cells(F=73.120,P<0.01).Conclusion NSCLCs transplantation exerts a protective effect against cerebral ischemia/reperfusion injury in rats by improving neuron injury.
