一起食物中毒事件中分离的阿邦尼沙门氏菌基因组信息

Genomic analysis of Salmonella Abani isolated from a food poisoning event

摘要 目的 分析一起食物中毒事件中分离的阿邦尼沙门氏菌的基因组信息,预测其致病机制和耐药机制。方法 对此次食物中毒事件中分离得到的阿邦尼沙门氏菌(Y202107)做全基因组测序,下机数据经过预处理后得到Clean Data,使用SOAP denovo (version 2.04),SPAdes,ABySS组装软件进行组装,并最终使用CISA软件进行整合,采用gapclose软件对初步组装结果进行优化和补洞,从最终的组装结果得到细菌完成图,与GO、KEGG、CAzy、CARD、VFDB数据库进行对比。结果 Y202107基因组大小为4 647 556 bp,预测共含4 435个基因, GC含量为53.32%。通过注释GO数据库,共比对到相关基因3 164个,分为3大类;通过对比KEGG数据库,共得到相关基因3 183个,参与24类代谢途径;通过对比CAZy数据库,共得到相关基因180个,分成6大类;通过对比CARD、VFDB数据库,得到4类耐药相关基因及沙门氏菌毒力岛SPI-1、SPI-2、SPI-3、SPI-4、SPI-5、SPI-6相关的致病性相关基因。结论 阿邦尼沙门氏菌的代谢过程共分为6大类,24小类;应重点关注因产生钝化酶而产生的耐药机制;与致病性最为相关的毒力因子是毒力岛。丰富了阿邦尼沙门氏菌基因组的数据库,为沙门氏菌病的疫苗研究提供的了基因层面的基础。 Objective To analyze the genomic information of Salmonella Abani isolated from a food poisoning event,and predict its pathogenic and drug resistance mechanisms.Methods Whole genome sequencing on the isolated Salmonella Abani(Y202107)from this food poisoning incident was performed,the offline data was preprocess to obtain clean data,which was assembled using SOAP denovo(version 2.04),SPAdes,ABySS assembly software,and finally integrated using CISA software.It was used gapclose software to optimize and fill in the preliminary assembly results,and obtain bacterial completion maps from the final assembly results,which were compared with GO,KEGG CAzy,CARD,and VFDB databases.Results The Genome size of Y202107 was 4647556 bp.It was predicted that there were 4435 genes,and the CC-content was 53.32%.By annotating the GO database,a total of 3164 related genes were identified and classified into 3 major categories.By comparing with the KEGG database,a total of 3183 related genes were identified,which were involved in 24 metabolic pathways;By comparing with CAZy database,a total of 180 related genes were identified and divided into 6 categories;By comparing with CARD and VFDB databases,four types of drug-resistance related genes and pathogenic related genes related to SPI-1、SPI-2、SPI-3、SPI-4、SPI-5、SPI-6of Salmonella pathogenicity island were obtained.Conclusions The metabolic process of Salmonella Abani can be divided into 6 categories and 24 subcategories;emphasis should be placed on the mechanisms of drug resistance caused by the production of inactivating enzymes;the most relevant virulence factor to pathogenicity is the pathogenicity island.the database of Salmonella Albani genome was enriched for providing a genetic foundation for vaccine research on Salmonella disease.
