人脐带间充质干细胞来源外泌体对大鼠卵巢早衰模型治疗效果的研究

摘要 目的探讨POF的治疗方法。方法将15只清洁级雌性SD大鼠随机分为A、B、C三组,各5只。A组注射同体积生理盐水,B、C组通过腹腔注射顺铂建立大鼠POF动物模型。模型建立后,C组注射huMSCs-exo。观察各组不同时点体重及血清中FSH、LH、E_(2)水平的变化。结果随时间的延长时间,A组大鼠体重呈增高趋势,B、C组呈递降趋势(P<0.05);造模完成后,B组E_(2)呈递降趋势,FSH呈递升趋势,LH呈先升后降趋势;C组E_(2)呈先降后升趋势,FSH、LH呈先升后降趋势(P<0.05)。结论HuMSCs-exo通过改善激素水平来修复大鼠卵巢功能,具有治疗POF的潜力。 Objective To explore the treatment of POF.Methods Fifteen clean grade female SD rats were randomly divided into three groups A,B and C with 5 rats in each group.Group A was injected with the same volume of normal saline,group B and group C were injected intraperitoneally with cisplatin to establish POF animal model.After the model was established,group C was injected with huMSCs-exo.The changes of body weight and serum levels of FSH,LH and E_(2)were observed in each group at different time points.Results With the extension of time,the body weight of rats in group A showed an increasing trend,while that in groups B and C showed a decreasing trend(P<0.05).After the molding was completed,E_(2)of group B showed a decreasing trend,FSH showed a progressive trend,LH showed a first increasing and then decreasing trend.In group C,E_(2)decreased first and then increased,FSH and LH increased first and then decreased(P<0.05).Conclusion HuMSCs-exo can repair the ovarian function of rats by improving hormone levels and has the potential to treat POF.
