防脱育发液对雄激素性脱毛大鼠毛囊及微环境的影响研究

Effects of Fangtuo Yufa Essence on the Hair Follicles and Microenvironment in Rats with Androgenetic Alopecia

摘要 目的:探讨防脱育发液对雄激素性脱毛大鼠皮肤毛囊及微环境的影响。方法:随机将45只SPF级SD大鼠分为空白组、对照组、模型组、米诺地尔组、防脱育发液组,采用皮下注射丙酸睾酮溶剂(5 mg/kg/d)的方法建立雄激素性脱毛大鼠模型。造模同时给药,1次/d,时间为35 d;空白组不用药,对照组和模型组给予0.2 mL生理盐水外涂,米诺地尔酊组予0.2 mL米诺地尔酊外涂,防脱育发液组予0.2 mL防脱育发液外涂。观察大鼠一般状态,运用HE染色、皮肤镜、Western blot手段观察大鼠皮肤毛囊密度数、病理学结构、局部微环境和肝细胞生长因子(HGF)表达的情况。结果:防脱育发液组大鼠毛发脱落少于模型组,病理学显示可增加毛囊密度数量(P<0.05),减轻大鼠毛囊和皮脂腺病变(P<0.05),明显升高HGF蛋白表达水平(P<0.05);皮肤镜下模型组毛发明显稀疏、油腻,较同期米诺地尔酊组、防脱育发液组黄点征明显,毛周褐色征偶见于第4-5周的米诺地尔酊组、防脱育发液组。结论:防脱育发液可促进雄激素性脱毛大鼠的毛发生长,其机制可能是增加毛囊密度数,减轻毛囊和皮脂腺病变,促进HGF蛋白表达以及改善毛囊微环境。 Objective:To investigate the effects of Fangtuo Yufa Essence on the skin follicles and their microenvironment of rats with androgenetic alopecia.Methods:Forty-five SPF SD Rats were randomLy divided into blank,control,model,minoxidil,and Fangtuo Yufa Essence groups.An androgenic alopecia rat model was established by subcutaneous injection of testosterone propionate solvent(5mg/kg/d).The model was administered once daily for 35 days,while the blank group received no medication.The control group and the model group received 0.2 mL of saline,the minoxidil group received 0.2 mL of minoxidil,and the Fangtuo Yufa Essence group received 0.2 mL of Fangtuo Yufa Essence.The general condition of rats was observed,and the number of hair follicle density,pathological structure,local microenvironment and expression of hepatocyte growth factor(HGF)were observed by HE staining,dermoscopy and Western blot.Results:The rats in the Fangtuo Yufa Essence group shed less hair than those in the model group,and the pathology confirmed that the number of hair follicles could be increased(P<0.05),reduced the hair follicles and sebaceous glands in rats could be reduced(P<0.05),and increased the expression level of HGF protein(P<0.05).dermoscopically,the model group had obvious hair thinning and greasy,and the yellow dots sign was obvious compared with the minoxidil group and Fangtuo Yufa Essence group in the same period,and the brown peripilar sign was occasionally seen in the minoxidil group and Fangtuo Yufa Essence group at week 4~5.Conclusion:Fangtuo Yufa Essence stimulates hair growth in rats with androgenetic alopecia,possibly by increasing follicle density,reducing follicle and sebaceous gland lesions,increasing the expression of HGF protein,and improving the microenvironment of the hair follicle.
