摘要
目的:通过数据挖掘“鸡血藤-海风藤”抗风湿的物质基础及作用机理。方法:通过TCMSP数据库获取“鸡血藤-海风藤”药对的成分,并以OB≥30%、DL≥0.18进行筛选,获取筛选后有效成分的基因靶点,在OMIM、TTD、CTD、Gene Cards等数据库中以“rheumatoid”为关键词进行疾病靶点检索,两者取交集,利用Cytoscape-v 3.8.2软件绘制网络图,利用R语言进行GO和KEGG富集分析,并分析相关数据。结果:共获得45个有效成分和24个共有关键靶点。通过药物-成分-疾病-靶点网络结果分析,其有效成分可能为木犀草素、芒柄花黄素、β-谷甾醇;关键靶点可能是PTGS2、PTGS1、MAPK14,主要富集于TH17细胞分化、PI3K-Art和TNF等信号通路。结论:“鸡血藤-海风藤”药对中的木犀草素、芒柄花黄素、β-谷甾醇等成分很可能通过PTGS2、PTGS1、MAPK14等靶点作用于TH17细胞分化、PI3K-Art和TNF等多条信号通路发挥抗风湿作用。
Objective:To explore the material basis and mechanism of anti-rheumatism of“Spatholobi Caulis-Piperis Kadsurae Caulis”through data.Methods:The components of the“Spatholobi Caulis-Piperis Kadsurae Caulis”drug pair were obtained through the TCMSP database,the OB≥30%and DL≥0.18 were screened to obtain the gene targets of the active ingredients after screening,and the disease targets were searched with“rheumatoid”as the keyword in OMIM,TTD,CTD,Gene Cards and other databases,and the two were taken as intersected.Cytoscape-v 3.8.2 software was used to draw network diagrams,and R language was used for GO and KEGG enrichment analysis,and related data were analyzed.Results:A total of 45 active ingredients and 24 common key targets were obtained.Through the analysis of drug-component-disease-target network results,the active ingredients may be:luteolin,formononetin,β-sitosterol;Key targets may be:PTGS2,PTGS1,MAPK14,mainly enriched in TH17 cell differentiation,PI3K-Art and TNF signaling pathways.Conclusion:Luteolin,formononetin,β-sitosterol and other components in the Spatholobi Caulis-Piperis Kadsurae Caulis drug pair are likely to exert anti-rheumatic effects on TH17 cell differentiation,PI3K-Art and TNF through targets such as PTGS2,PTGS1 and MAPK14.
