基于网络通路调控食管鳞状细胞癌发生的分子机制

Molecular mechanism of regulation of esophageal squamous cell carcinoma based on network pathway

摘要 目的:探讨食管鳞状细胞癌(esophageal squamous cell carcinoma,ESCC)中circSSPO/microRNA-6820-5p(miR-6820-5p)网络通路调控ESCC增殖、迁移和侵袭的可能分子机制。方法:对收集得到的ESCC组织样本进行RNA提取和全转录组测序和差异分析,使用流式细胞术检测细胞的凋亡水平,平板克隆检测细胞的增殖情况,Transwell迁移和侵袭实验检测细胞的迁移与侵袭情况。结果:生物信息学预测和RNA测序结果筛选得出circSSPO和miR-6820-5p与ESCC的进展相关,在ESCC中,miR-6820-5p低表达,circSSPO高表达。而且通过体外细胞实验进一步证明circSSPO可以通过抑制miR-6820-5p从而诱导ESCC细胞致癌特性。结论:circSSPO是一种在ESCC组织中丰富表达的致癌circRNA,并且circSSPO/miR-6820-5p网络通路与ESCC的增殖、迁移和侵袭有关。 OBJECTIVE To investigate the possible molecular mechanism of circSSPO/microRNA-6820-5p(miR-6820-5p)in esophageal squamous cell carcinoma(ESCC).METHODS The collected ESCC tissue samples were subjected to RNA extraction,whole transcriptome sequencing and differential analysis.The flow cytometry,the plate cloning and Transwell migration and invasion assay was used to detect the apoptosis level,the proliferation,and the migration and invasion of cells.RESULTS According to bioinformatics prediction and RNA sequencing results,circSSPO and miR-6820-5p are associated with ESCC development.In ESCC,the expression of miR-6820-5p was low,while the expression of circSSPO was high.The vitro cell experiments further demonstrated that circSSPO competitively inhibited miR-6820-5p induced oncogenic properties of ESCC cells.CONCLUSION CircSSPO is an oncogenic circRNA abundantly expressed in ESCC tissues,and the circSSPO/miR-6820-5p network pathway is related to the proliferation,migration and invasion of ESCC.
