膜片钳技术在生物医学研究中的联合应用及其发展趋势

摘要 膜片钳技术是研究离子通道电活动的“金标准”,不仅用于研究生理和病理条件下细胞膜离子通道电生理特性和膜电位的变化,还广泛应用于分析药物作用离子通道机制、优化化合物结构以及评价药物毒性等方面。目前,膜片钳技术已成为现代细胞电生理学研究的常规方法,广泛应用于神经科学、心血管科学、药理学、细胞生物学等领域并取得了丰硕的研究成果。近年来,随着基因组学、光遗传学、质谱技术和芯片技术等的不断涌现和日益完善,这些新技术与膜片钳技术的联合应用极大促进了以离子通道为靶点的药物研发和对疾病发病机制的深入理解,进一步拓宽了生命现象的研究维度,促进了生命科学研究的发展。 Patch clamp technology is the"gold standard"for studying the electrical activity of ion channels.It is not only used to study the electrophysiological characteristics and membrane potential changes of cell membrane ion channel under physiological and pathological conditions,but also widely used to analyze the mechanism of drug action on ion channels,optimize the structure of compounds,and evaluate drug toxicity.At present,patch clamp technology has become a conventional method of modern cell electrophysiology research,which has been widely used in neuroscience,cardiovascular science,pharmacology,cell biology and other fields,and has achieved fruitful research results.In recent years,with the continuous emergence and improvement of genomics,optogenetics,mass spectrometry and chip technology,the joint application of these new technologies and patch clamp technology has greatly promoted drug development targeting ion channels and in-depth understanding of the pathogenesis,further widened the research dimension of life phenomena,and promoted the development of life science research.
