摘要
目的分析非小细胞肺癌(NSCLC)患者血清微小核糖核酸-92a(miR-92a)、微小核糖核酸-143(miR-143)、微小核糖核酸-196b(miR-196b)变化及其临床意义。方法选取70例NSCLC患者作为NSCLC组,同期选取70例肺部良性病变患者为对照组。采用实时定量RT-PCR检测血清miR-92a、miR-143、miR-196b表达量;采用单因素生存分析Kaplan-Meier绘制3年生存曲线,采用Pearson相关性分析NSCLC组和对照组血清miR-92a、miR-143、miR-196b水平相关性。结果NSCLC患者中血清miR-92a、miR-196b表达水平显著高于对照组(P<0.05),血清miR-143表达显著低于对照组(P<0.05);生存分析显示,血清中miR-92a、miR-196b低表达患者3年总生存率均显著高于miR-92a、miR-196b高表达患者(P<0.05);血清miR-143高表达患者3年总生存率均高于低表达组(P<0.05)。Pearson相关性分析显示,NSCLC患者中血清miR-92a与miR-196b表达呈正相关性(γ=0.425,P<0.05),miR-143与miR-92a、miR-196b表达呈负相关(γ=-0.376,-0.356,P<0.05)。结论血清miR-92a、miR-196b在NSCLC患者中表达升高,血清miR-143表达降低,血清miR-92a、miR-143、miR-196b水平变化可作为NSCLC临床病情评估和预后的重要依据。
Objective To analyze the changes and clinical significance of serum microRNA-92a(miR-92a),microRNA-143(miR-143)and microRNA-196b(miR-196b)in patients with non-small cell lung cancer(NSCLC).Methods A total of 70 patients with NSCLC were enrolled as NSCLC group,while 70 patients with benign lung lesions during the same period were enrolled as the control group.The expressions levels of serum miR-92a,miR-143 and miR-196b were detected by real-time quantitative RT-PCR.The 3-year survival curves were drawn by univariate survival Kaplan-Meier analysis.The correlation of serum miR-92a,miR-143 and miR-196b in NSCLC group and the control group was analyzed by Pearson correlation analysis.Results The expression levels of serum miR-92a and miR-196b in NSCLC group were significantly higher than those in the control group(P<0.05),while level of serum miR-143 was significantly lower than that in the control group(P<0.05).The survival analysis showed that 3-year overall survival rates in patients with low expressions of miR-92a and miR-196b were significantly higher than those with high expressions,respectively(P<0.05).The 3-year overall survival rate in patients with high expression of miR-143 was higher than that with low expression(P<0.05).Pearson correlation analysis showed that serum miR-92a was positively correlated with miR-196b(γ=0.425,P<0.05),and miR-143 was negatively correlated with miR-92a and miR-196b(γ=-0.376,-0.356,P<0.05).Conclusion The expressions of serum miR-92a and miR-196b are up-regulated,while expression of serum miR-143 is down-regulated in NSCLC patients.The changes of serum miR-92a,miR-143 and miR-196b can be applied as important basis to clinically evaluate conditions and prognosis of NSCLC.
