基于网络药理学探讨四妙丸治疗腰椎间盘突出症的作用机制

摘要 目的:采用网络药理学的研究方法,对四妙丸用于治疗腰椎间盘突出症(LDH)的药理作用以及机制问题进行研究和探讨。方法:借助TCMSP平台筛选出四妙丸活性药物成分及靶标预测。从OMIM数据库、GeneCards数据库、Drugbank数据库中检索出疾病靶标。采用R软件确定了四妙丸-LDH的交集靶点并利用STRING网站进行蛋白互作。利用Cytoscape软件构建了药物成分-交集靶点网络。最后在R软件将交集靶标进行富集分析。结果:分析研究显示共挖掘到了四妙丸中活性药物成分75种及预测靶点2902个、399个LDH疾病相关靶点,取交集后得到四妙丸-LDH交集靶点45个,分析得到MAPK14、MAPK1、IL6、ESR1、FOS和JUN可能为核心靶点,汉黄芩素、槲皮素可能为四妙丸治疗LDH的关键活性化合物。GO富集分析得到GO条目2004个,KEGG分析得到AGE-RAGE信号通路、TNF信号通路、IL-17信号通路等可能为作用通路。结论:四妙丸中有多个对LDH有抑制作用的药效成分,多成分多靶标共同作用是其作用机制。 Objective The pharmacological effects and mechanism of Simiao Pill(SMW)in the treatment of lumbar disc herniation(LDH)were studied and discussed using the pharmacological research method of network platform.Method With the help of the data-base and analysis platform of traditional Chinese medicine system(TCMSP),the active drug components of SMW were screened and their targets were predicted.Retrieve LDH disease targets from the database.The Venn atlas was constructed by R software,and the target of SMW in treating LDH was determined.The component disease analysis network was constructed by using the Cytoscape soft-ware.At the same time,the STRING website was used to form an interaction network between proteins,and the core target was deter-mined in the Cytoscape software.The target is enriched and analyzed in R software to determine the biological function and core path-way of SMW in treating LDH.Results Analysis and research showed that 75 kinds of active drug ingredients,2902 predicted targets and 399 LDH disease related targets in SMW were discovered.After the intersection,45 SMW LDH intersection targets were obtained.Topological analysis showed that MAPK14,MAPK1,IL-6,ESR1,FOS,JUN may be the core target.Hanbaicalin and quercetin may be the key active compounds of SMW in the treatment of LDH.There are 2004 GO entries obtained by GO enrichment analysis.KEGG analysis revealed that the AGE-RAGE signaling pathway,TNF signaling pathway,IL-17 signaling pathway,and other potential path-ways may be involved.Conclusion In SMW,there are several active ingredients that have inhibitory effect on LDH,and the mecha-nism of action is the joint action of multiple components and multiple targets.
