摘要
目的:探讨敲降长链非编码RNA00467(LINC00467)调控代谢重编程对肺腺癌(LAD)增殖及迁移的影响。方法:qRT-PCR验证H1299、A549细胞和正常支气管上皮细胞(16HBE)中LINC00467的表达情况。H1299、A549细胞中使用慢病毒(shRNA)敲降LINC00467,通过平板克隆、Transwell、流式细胞术检测细胞增殖、迁移和凋亡率。使用慢病毒敲降的H1299、A549细胞验证己糖激酶2(Hexokinase2)和葡萄糖转运体1(Glucose transporter1)的表达含量。使用乳酸试剂盒和ATP试剂盒检测被慢病毒敲降LINC00467的H1299、A549细胞的乳酸含量和ATP含量。结果:LINC00467在H1299和A549细胞中的表达上调(P<0.001);在H1299和A549细胞中敲降LINC00467后能明显抑制细胞的增殖和迁移能力,并促进细胞的凋亡。Hexokinase2和Glucose transporter1在被敲降LINC00467的H1299和A549细胞中表达降低(P<0.001)。被敲降后的H1299和A549细胞的乳酸含量和ATP含量与对照组相比降低(P<0.001)。结论:敲降LINC00467可以降低LAD细胞的能量代谢水平,从而影响LAD增殖及迁移能力。
Objective:To investigate the knockdown of long-stranded non-coding RNA00467 that regulates metabolic reprogramming and thus affects lung adenocarcinoma proliferation and migration.Methods:qRT-PCR was performed to verify the expression of LINC00467 in H1299,A549 cells and normal bronchial epithelial cells(16HBE).Lentiviral knockdown of LINC00467 in H1299,A549 cells was used to detect cell proliferation,migration and apoptosis rates by plate cloning,Transwell and flow cytometry.The expression levels of hexokinase 2 and glucose transporter 1 were verified using lentiviral knockdown of H1299,A549 cells.The lactate content and ATP content of H1299,A549 cells knocked down by LINC00467 lentivirus were measured using the lactate kit and ATP kit.Results:LINC00467 expression was significantly upregulated in H1299 and A549 cells(P<0.001);Knockdown of LINC00467 in H1299 and A549 cells significantly inhibited cell proliferation and migration ability,and promoted apoptosis;The expression of hexokinase 2 and glucose transporter 1 was significantly reduced in H1299 and A549 cells knocked down by LINC00467(P<0.001);The lactate content and ATP content of H1299 and A549 cells knocked down by LINC00467 were significantly decreased compared with the control group(P<0.001).Conclusion:Knockdown of LINC00467 can reduce the energy metabolism level of lung adenocarcinoma cells,thus affecting the proliferation and migration ability of lung adenocarcinoma.
