Chemical Analysis of Sorghum Husk by UPLC-Q Exactive Plus-MS in Vitro Screening of Xanthine Oxidase Inhibitory Activity and Composition

摘要 采用高效液相色谱串联四极杆静电场轨道阱质谱对高粱壳中的化学成分进行分析鉴定,并通过高效液相色谱法筛选抑制黄嘌呤氧化酶活性成分。该实验使用Hypersil GOLD C_(18)色谱柱(2.1 nm×30 nm,3μm),以0.1%甲酸水(A)-乙腈(B)进行梯度洗脱,流速1.0 m L/min,进样量10μL。质谱分析采用ESI离子源,在正离子模式下采集数据,利用Xcalibur 4.1软件结合数据库对所得数据进行分析,共鉴定出27种化学成分。对刺槐素、葛根素、芹菜素、根皮苷、异鼠李素和3,3’,4’,7-四羟基黄酮6种化学成分进行活性筛选,发现芹菜素和3,3’,4’,7-四羟基黄酮对黄嘌呤氧化酶(XO)表现出较强的抑制活性,IC_(50)分别为18.11和34.59μmol/L。 The chemical components in sorghum husk are analyzed and identified by high performance liquid chromatography tandem with quadrupole electrostatic field orbital trap mass spectrometry and screened for inhibition of xanthine oxidase activity components by high performance liquid chromatography.The experiment is performed on a Hypersil GOLD C_(18) column(2.1 nm×30 nm,3μm)with a gradient elution of 0.1% formic acid water(A)-acetonitrile(B)at a flow rate of 1.0 mL/min and a sample volume of 10μL.The mass spectrometry analysis is performed using an ESI ion source and data are collected in positive ion mode.The data obtained are analyzed using Xcalibur 4.1 software in combination with a database,and a total of 27 chemical components are identified.Six chemical components,namely acacetin,puerarin,apigenin,phlorizin,isorhamnetin and fisetin,are screened for activity,and apigenin and fisetin are found to exhibit strong inhibitory activity on xanthine oxidase(XO)with IC_(50) of 18.11 and 34.59μmol/L,respectively.
