探究三七多糖对COPD大鼠肺部巨噬细胞极化及炎症影响 被引量:5

Effect of Panax Notoginseng Polysaccharide on Pulmonary Inflammation and Macrophage Polarization in Chronic Obstructive Pulmonary Disease Rats

摘要 目的探讨三七多糖(panaxnotoginseng poiysaccharides,PNP)通过调控慢性阻塞性肺疾病(chronic obstructive pulmonary disease,COPD)大鼠肺部巨噬细胞极化缓解肺部炎症的可能机制。方法采用烟熏联合气管滴注脂多糖的方法复制COPD大鼠模型,造模成功后以灌胃的方式给予三七多糖干预。EMMS用力肺功能系统检测大鼠的肺功能;HE染色及Masson染色检测肺组织炎症浸润情况及胶原沉积情况;ELISA法检测肺组织匀浆中TNF-α、IL-1β、IL-6、IL-8的表达水平;流式细胞技术及免疫荧光检测肺组织中M1/M2型巨噬细胞的表达水平。结果与空白组相比,模型组大鼠肺功能下降,且差异具有统计学意义(P<0.05);肺组织炎性细胞大量浸润、胶原沉积增加;肺组织匀浆中TNF-α、IL-1β、IL-6、IL-8的表达升高,且差异具有统计学意义(P<0.05);M1型巨噬细胞的表达水平上升,M2型巨噬细胞的表达下降,且差异具有统计学意义(P<0.05);与模型组相比,给药组的肺功能上升,且差异具有统计学意义(P<0.05);肺组织炎性细胞浸润减少、胶原沉积情况有所改善;肺组织匀浆中TNF-α、IL-1β、IL-6、IL-8的表达降低,且差异具有统计学意义(P<0.05);M1型巨噬细胞的表达水平降低,M2型巨噬细胞的表达上升,且差异具有统计学意义(P<0.05)。结论三七多糖可以改善COPD大鼠的肺功能,降低肺部M1型巨噬细胞及炎症因子的表达水平,提高M2型巨噬细胞的表达水平,从而缓解肺部炎症,减轻COPD的病变进程。 Objective To investigate the possible mechanism of panax notoginseng poiysaccharides(PNP)relieving pulmonary inflammation by regulating pulmonary macrophage polarization in chronic obstructive pulmonary disease(COPD)rats.Methods The rat model of COPD was established by smoking combined with intratracheal instillation of lipopolysaccharide.EMMS forced pulmonary function system was used to detect the pulmonary function of rats;HE staining and Masson staining were used to detect the inflammatory infiltration and collagen deposition in lung tissues;ELISA was used to detect the expression levels of TNF-α,IL-1β,IL-6,and IL-8 in lung homogenates;flow cytometry and immunofluorescence were used to detect the expression levels of M1/M2 macrophages in lung tissues.Results Compared with the blank group,the pulmonary function of rats in the model group decreased,and the difference was statistically significant(P<0.05);a large number of inflammatory cells infiltrated and collagen deposition increased in the lung tissue;the expression of TNF-α,IL-1β,IL-6,and IL-8 increased in the lung homogenate,and the difference was statistically significant(P<0.05);the expression level of M1 macrophages increased,and the expression of M2 macrophages decreased,and the difference was statistically significant(P<0.05);compared with the model group,the pulmonary function of the treated group increased,and the difference was statistically significant(P<0.05);the inflammatory cell infiltration decreased and collagen deposition in the lung tissue improved;the expression of TNF-α,IL-1β,IL-6,and IL-8 decreased in the lung homogenate,and the difference was statistically significant(P<0.05);the expression level of M1 macrophages decreased,and the expression level of M2 macrophages increased,and the difference was statistically significant(P<0.05).Conclusion Panax notoginseng polysaccharide can improve the pulmonary function,decrease the expression of M1 macrophages and inflammatory factors,and increase the expression of M2 macrophages and anti-inflammatory factors in COPD rats,so as to relieve pulmonary inflammation and reduce the pathological process of COPD.
