摘要
为了获得对花椒根腐病病原真菌腐皮镰孢菌有拮抗效果的贝莱斯芽孢杆菌T-1的最佳培养环境,通过单因素试验筛选出对贝莱斯芽孢杆菌T-1菌体浓度影响较大的四个因素:pH值、温度、接种量、转速.采用Box-Bennie中心组合设计试验,对其发酵培养参数进行优化,以提高其菌浓度.结果表明:优化后的最佳发酵条件为:pH7.0、温度30℃、接种量为4%、转速181 r/min,测得的OD600值为1.905,与模型预测值基本相符.优化后的菌落数可达83.6×10^(8)CFU/mL,与优化前相比提高了19.08倍.
Bacillus velezensis T-1 is an antagonistic bacterium screened in previous experiments,which has good antagonistic ef-fects on the pathogenic fungus Fusarium solani of Zanthoxylum bungeanum root rot.Four factors that have a significant impact on the concentration of Bacillus velezensis T-1 were screened through single factor experiments:pH value,temperature,inoculation amount,and rotational speed.Using the Box-Benie central combination design experiment,the fermentation and cultivation pa-rameters were optimized to increase the bacterial concentration.The results show that the optimal fermentation conditions for this strain after optimization are pH 7.0,temperature 30°C,inoculation amount 4%,and rotational speed 181 r/min.The measured OD600 is 1.905,which is basically consistent with the predicted value of the model.The optimized colony count can reach 83.6×10^(8)CFU/mL,with an increase of 19.08 times compared to that before optimization.
