血平片抗大鼠子宫肌瘤作用研究

Effect of XuePing Tablets Against Hysteromyoma in Rats

目的探究血平片与抑那通分别单独使用与联用时抗子宫肌瘤的作用。方法将80只成熟未孕的雌性SD大鼠随机选取10只作为空白组,剩余大鼠通过雌孕激素双重造模法建立大鼠子宫肌瘤模型,分为模型组、血平片中、高剂量组、抑那通低、高剂量组、中、高剂量联合组,连续给药4周。记录大鼠体重和子宫、卵巢湿质量,ELISA法测定血清中孕酮(P)、雌二醇(E 2)含量,HE染色后观察子宫形态变化,采用IHC,检测组织中ER蛋白、PR蛋白的表达水平。结果模型组与空白组大鼠的体重、子宫及卵巢的湿重与系数、平滑肌厚度、激素水平和ER、PR表达有明显差异,高剂量血平片给药(756 mg·kg^(-1))对子宫平滑肌增生有一定的抑制作用,中剂量联合给药(378 mg·kg^(-1)血平片、0.5 mg·kg^(-1)抑那通)、高剂量联合给药(756 mg·kg^(-1)血平片、0.5 mg·kg^(-1)抑那通)、低剂量抑那通给药(0.5 mg·kg^(-1))和高剂量抑那通给药(1 mg·kg^(-1))可以使子宫平滑肌厚度降低到空白组水平。结论血平片通过抑制子宫平滑肌细胞ER、PR表达从而改善子宫肌瘤模型大鼠的症状。血平片与抑那通联合给药可以使平滑肌层厚度降低到正常水平且效果优于单独使用。

OBJECTIVE To explore the effect of XuePing tablets and Enantone against uterine hysteromyoma.METHODS Ten out of 80 mature unpregnant female SD rats were randomly selected as blank group,and the remaining rats established the rat uterine fibroids model by estrogen progesterone dual molding method,which was divided into model group,XuePing tablets medium and high dose group,low,Enantone high dose group,medium and high dose combination group,and continuously administered for 4 weeks.The body weight and wet mass of uterus and ovary were recorded,and the serum contents of progesterone(P)and estradiol(E 2)were determined by ELISA.The morphological changes were observed after HE staining.IHC was used to detect the expression level of ER protein and PR protein in tissues.RESULTS There were significant differences in wet weight and coefficient,uterus and ovary,smooth muscle thickness,hormone levels and ER and PR expression between blank and moded rats,High-dose XuePing tablets administration(756 mg·kg^(-1))had some inhibitory effect on uterine smooth muscle hyperplasia,Medium-dose combination(378 mg·kg^(-1) XuePing tablets,0.5 mg·kg^(-1) Enantone),high-dose combination(756 mg·kg^(-1) XuePing tablets,0.5 mg·kg^(-1) Enantone),low-dose Enantone(0.5 mg·kg^(-1))and high-dose Enantone(1 mg·kg^(-1))reduced uterine smooth muscle thickness to blank group level.CONCLUSION XuePing tablets improved the symptoms of rats in the uterine myoma model by inhibiting ER and PR expression in uterine smooth muscle cells.Combined administration of XuePing tablets and Enantone has reduced the smooth muscle layer thickness to normal levels and works better than that alone.
