摘要
目的研究肾康灵胶囊联合泼尼松对阿霉素肾病(Adriamycin nephropathy,AN)大鼠的肾脏保护作用,初步探索肾康灵胶囊在慢性肾病激素治疗中发挥的药效学作用。方法雄性SD大鼠随机分为正常组、模型组、泼尼松组和联合治疗组(泼尼松+肾康灵胶囊)。大鼠尾静脉注射阿霉素制备AN模型并进行药物治疗,检测大鼠血清ALB、BUN、Cr、CHOL和TG等指标及24 h尿蛋白量水平,HE染色观察肾脏组织形态学变化。结果与模型组相比,泼尼松组和联合治疗组可降低大鼠血清BUN、Cr、CHOL、TG水平,升高ALB水平,HE染色结果显示可减少肾脏组织损伤。与泼尼松组相比,联合治疗组可升高AN大鼠血清ALB水平,显著降低AN大鼠24 h尿蛋白量及血清CHOL、TG水平。结论与单纯泼尼松治疗相比,肾康灵胶囊联合泼尼松可进一步减少AN大鼠的肾脏损伤,显著改善AN大鼠的血脂代谢异常。
OBJECTIVE To investigate the renal protective effect of prednisone combined with ShenKangLing capsules on adriamycin nephropathy(AN)rats,and to initially explore the pharmacodynamic role of ShenKangLing capsules in the hormone therapy of chronic kidney disease.METHODS Male SD rats were randomly divided into 4 groups:Control group,Model group,Prednisone group,Prednisone+ShenKangLing capsules group(Combination treatment group).AN model was prepared by intravenous injection of adriamycin into the tail of rats and treated with drugs,serum ALB,BUN,Cr,CHOL,TG and 24 h urinary protein levels of rats were detected,and renal histomorphology was observed by HE staining.RESULTS Compared with Model group,the levels of BUN,Cr,CHOL and TG were decreased and ALB was increased in Prednisone group and combination treatment group.HE staining showed that Prednisone group and combination treatment group could reduce renal tissue injury.Compared with Prednisone group,combination treatment group could increase serum ALB level and significantly reduce the 24 h urine protein volume and serum CHOL,TG levels of AN rats.CONCLUSION Compared to treatment with prednisone alone,prednisone combined with ShenKangLing capsules can further reduce kidney injury and significantly ameliorate dyslipidemia in AN rats.
