赤桉蜂蜜抗炎作用的初步研究

摘要 目的:探究赤桉蜂蜜(Jarrah Honey,JH)对于脂多糖(Lipopolysaccharide,LPS)诱导RAW264.7细胞炎症反应的影响。方法:将RAW264.7随机分为空白对照组、LPS组、Honey组、JH组,用CCK-8法检测细胞活力,筛选出合适的给药浓度,LPS诱导RAW264.7构建体外炎症模型,通过ELISA测定炎症因子白细胞介素-6(IL-6)、IL-1β、α肿瘤坏死因子(TNF-α)的浓度。结果:炎症刺激后,模型组中IL-6、IL-1β和TNF-α的浓度明显增加。给予JH后,显著提高了LPS诱导炎症刺激后RAW264.7细胞活力,IL-6、IL-1β和TNF-α的分泌量减少。在40μg/mL时,与Honey组相比,JH组IL-6、IL-1β和TNF-α分泌量显著减少。结论:JH可以增强细胞抗炎能力降低炎症影响,对LPS诱导炎症损伤后的RAW264.7有较好的保护效果。 Objective:To investigate the effect of Jarrah Honey(JH)on LPS-induced inflammatory response in RAW264.7 cells.Methods:RAW264.7 is randomly divided into blank control group,LPS group,Honey group and JH group,cell viability is detected by CCK-8 method,the appropriate administration concentration is screened,LPS induced RAW264.7 to construct an in vitro inflammatory model,and the concentrations of inflammatory factors interleukins-6(IL-6),IL-1βand tumor necrosis factor-α(TNF-α)are determined by ELISA.Results:After inflammatory stimulation,the concentrations of IL-6,IL-1βand TNF-αin the model group increased significantly.After JH administration,the viability of RAW264.7 cells is significantly increased after LPS-induced inflammatory stimulation,and the secretion of IL-6,IL-1βand TNF-αdecreased.At 40μg/mL,IL-6,IL-1βand TNF-αsecretions are significantly reduced in the JH group compared with the Honey group.Conclusion:JH can enhance cell anti-inflammatory ability and reduce inflammatory effects,and has a good protective effect on RAW264.7 after LPSinduced inflammatory injury.
