摘要
目的:判断珍珠岩细菌内毒素检查溶液的获取方式对使用鲎试剂(凝胶法)检测细菌内毒素有无干扰作用。方法:采用细菌内毒素凝胶法检测助滤剂珍珠岩的细菌内毒素,确认珍珠岩被高温干烤和未经高温干烤所得到的细菌内毒素检查溶液对内毒素检测的干扰。结果:样品前处理采用180℃干烤3 h与未干烤相比较均对检测方法无干扰,检测珍珠岩细菌内毒素的鲎试剂选择标示灵敏度为0.06 EU/mL。珍珠岩细菌内毒素检查溶液的获取方式:将供试品加细菌内毒素检查用水将供试品稀释8倍混匀4 000 r/min离心10 min提取后,选择经过验证不含内毒素和干扰因子的稀释剂Ⅱ与供试品按照1∶1的比例进行稀释,能有效避免珍珠岩对细菌内毒素检查的干扰。结论:通过采用稀释剂Ⅱ对样品进行稀释及中和,珍珠岩能够采用鲎试剂检测细菌内毒素。
Objective:Determine whether the method of obtaining perlite bacterial endotoxin test solution interferes with the use of limulus reagent(gel method)to detect bacterial endotoxin.Methods:The content of bacterial endotoxins in Perlite,the main filter aid for production,is determined by the limit test method of bacterial endotoxins.Results:There is no difference between the samples pre-treated by drying at 180℃for 3 h and non dry.The sensitivity of limulus reagent to detect bacterial endotoxin in perlite is 0.06 EU/mL.The method of obtaining bacterial endotoxin test solution for perlite:dilute the test sample by 8 times with water for bacterial endotoxin test,mix well at 4000 r/min,and centrifuge for 10 minutes to extract.Then,select diluentⅡthat has been verified to be free of endotoxin and interference factors and dilute it with the test sample in a 1∶1 ratio,which can effectively avoid interference of perlite with bacterial endotoxin test.Conclusion:By using diluent II to dilute and neutralize the sample,perlite can be used to detect bacterial endotoxins with limulus reagent.
