基线等比增减设计结合熵权TOPSIS法优选百合生地黄抗焦虑抑郁最佳配比 被引量:2

Optimization of the best ratio of Lily and Rehmannia for anxiolytic and antidepressant effect based on increase-decrease baseline geometric proportion design combined with entropy weight TOPSIS method

摘要 目的探索百合-生地黄抗焦虑抑郁的最佳配比。方法采用基线等比增减设计法设计百合生地黄不同配伍比例,利用经典行为学测定方法(高架十字迷宫实验、旷场实验、强迫游泳实验、悬尾试验)对百合-生地黄不同配比进行抗焦虑抑郁药效比较,结合高效液相色谱法对百合-生地黄不同配比药液中的王百合苷A、B、C、毛蕊花糖苷、异毛蕊花糖苷溶出量进行比较,采用熵权法确定各评价指标权重,结合TOPSIS法优选百合生地黄最佳配比。结果百合生地黄不同配比抗焦虑抑郁效果及有效成分溶出均存在显著差异,其中最优配比为5∶5,表明该比例最能发挥百合和生地黄的协同增效作用,是百合生地黄抗焦虑抑郁的最佳配伍比例。结论熵权TOPSIS法能够准确、客观地反映百合生地黄不同配比的差异,可为中药配伍研究提供新方法。 Objective To explore the optimal ratio of Lilii Bulbus(LB)and Rehmanniae Radix(RR)for antidepressant and anx-iolytic.Methods Different ratios of LB and RR were designed with increase-decrease baseline geometric proportion design,ele-vated plus maze test,open field test,forced swimming test,and tail suspension test were used to evaluate the anxiolytic and anti-depressant effect.High performance liquid chromatography was used to measure the level of regaloside A,B,C,acteoside,and isoacteoside.The entropy weight method was used to determine the weight of each evaluation index.the best combination of LB and RR was selected by TOPSIS method.Results In different ratios of LB and RR,antidepressant and anxiolytic effects and dis-solution of active ingredients differed significantly.The optimal ratio was 5:5,as a result,LB and RR could best exert their syn-ergistic effects in this proportion.Conclusion Based on increase-decrease baseline geometric proportion design and the entropy weight TOPSIS method,the proportions of LB and RR are optimized accurately and objectively,and this method provide new new insights for the research of Chinese medicine compatibility.
