山西省部分地区猪源不动杆菌分子鉴定及其耐药性分析

Molecular identification and drug resistance analysis of Acinetobacter from pigs in some areas of Shanxi Province

摘要 为了解2019年山西省猪源不动杆菌的流行情况、菌种类型及其耐药性情况,本研究无菌采集山西省11个地市20个屠宰场的521份猪肺组织,并分离到30株猪源不动杆菌。通过不动杆菌的bla_(OXA-51-like)、rpoB、gyrB基因PCR扩增、测序、同源性分析对分离的30株猪源不动杆菌进行了菌种鉴定,并对鉴定出的不动杆菌进行了药敏试验、耐药性分析和耐药基因的检测。结果显示:30株猪源不动杆菌有2株鉴定为鲍曼不动杆菌(A.baumannii)、28株为非鲍曼不动杆菌。其中,28株非鲍曼不动杆菌有6株为A.lwoffii、10株为A.towneri、5株为A.junii、3株为A.ursingii,4株未能确定菌种;鉴定的26株猪源不动杆菌对苯唑西林、复方新诺明的耐药率较高,分别为76.9%和46.1%,且对3种及3种以上药物耐药的菌株占42.3%;耐药基因bla_(TEM)、sul-2基因检出率较高,分别为57.7%和46.2%。结果表明,山西省猪源不动杆菌鉴定出5个种,存在鲍曼不动杆菌,但以非鲍曼不动杆菌为主,其中A.towneri最多;山西猪源不动杆菌对苯唑西林、复方新诺明耐药性很强,并存在多重耐药性,部分耐药基因检出率较高。 In order to investigate the prevalence,strains type and drug resistance of Acinetobacter isolated from pig in Shanxi Province in 2019,30 strains of Acinetobacter isolated from 521 pig lung tissues in 11 cities of Shanxi province were identifiedby polymerase chain reaction,sequencing analysis,and sequence alignment analysis of bla_(oxa-51-like),rpoB and gyrB genes of Acinetobacter,and the drug resistance of the identified Acinetobacter was detected and analyzed.The results showed that two strains were identified as Acinetobacter baumannii and 28 strains were identified as non-Acinetobacter baumannii strains,among them,six strains were identified as A.lwoffii,ten strains were identified as A.towneri,five strains were identified as A.junii and three strains were identified as A.ursingii,but the remaining 4 strains were not successfully identified.The drug resistance rates of 26 Acinetobacter strains to oxacillin and cotrimoxazole were higher(76.9%and 46.1%,respectively),and the proportion of 26 Acinetobacter strains resistant to three or more drugs was 42.3%.The detection rates of bla_(TEM)and sul-2 genes were 57.7%and 46.2%,respectively.These data demonstrated that five species of Acinetobacter were identified in Shanxi and A.baumannii was present,but the non-Acinetobacter baumannii was predominant,including the most numerous of A.towneri strains.Acinetobacter from pigs in Shanxi Province has strong resistance to oxacillin and cotrimoxazole,and has multi-drug resistance,the detection rate of some drug resistance genes was high.
