盐诱导激酶3基因表达在人类肿瘤中的预后与免疫浸润相关性分析

Prognostic and Immune Infiltration Correlation Analysis of Salt-induced Kinase 3 Gene Expression in Human Tumors

摘要 目的:通过对盐诱导激酶3(SIK3)在人类肿瘤中的泛癌化分析,探讨SIK3基因在不同肿瘤中的表达和预后,寻找肿瘤免疫治疗的潜在靶点和肿瘤生物标志物。方法:通过TIMER数据库检索SIK3基因和蛋白在正常组织与肿瘤组织的的表达差异,使用cBioportal工具研究SIK3基因在各肿瘤中的变异情况,进行Kaplan-Meier生存曲线分析以评估泛癌化预后价值,根据EPIC、MCPCOUNTER、TIDE算法进行TGCA中SIK3与肿瘤相关成纤维细胞浸润相关性分析。结果:SIK3在膀胱尿路上皮癌(BLCA)、乳腺癌(BRCA)、结肠腺癌(COAD)等13种癌症中低表达。通过分析肿瘤微环境中SIK3表达与免疫浸润T细胞的关系,发现COAD、头颈鳞状细胞癌(HNSC)、HPV感染的头颈部鳞癌(HNSC-HPV+)、胰腺癌(PAAD)、直肠腺癌(READ)、胃癌(STAD)、睾丸生殖细胞瘤(TGCT)、胸腺癌(THYM)中SIK3表达水平与癌相关成纤维细胞浸润水平呈正相关,脑低级别胶质瘤(LGG)中SIK3表达水平与癌相关成纤维细胞浸润水平呈负相关。泛癌生存分析表明,SIK3高表达在肾上腺皮质癌(ACC)预后较差,SIK3低表达在肾透明细胞癌(KIRC)、LGG、间皮瘤(MESO)、PAAD、甲状腺癌(THCA)预后较差。结论:这些发现表明SIK3基因在肿瘤免疫微环境中起重要作用,并且确定了SIK3基因在多种癌症中具有预后价值,推测SIK3基因是癌症免疫疗法和肿瘤生物标志物的潜在靶点。 Objective:By analyzing salt-induced kinase 3(SIK3)pan-cancer in human tumors,to explore the expression and prognosis of SIK3 gene in different tumors,and to search for potential targets and tumor biomarkers for tumor immunotherapy.Methods:Differences in the expression of SIK3 gene and protein in normal and tumor tissues were retrieved from the TIMER database.The cBioportal tool was used to study SIK3 gene variants in each tumor,and Kaplan-Meier survival curve analysis was performed to assess the prognostic value of pan-cancer.Correlation analysis between SIK3 and tumor-associated fibroblast infiltration in TGCA was performed according to EPIC,MCPCOUNTER,and TIDE algorithms.Results:SIK3 was lowly expressed in 13 cancers,including bladder uroepithelial carcinoma(BLCA),breast cancer(BRCA),and colon adenocarcinoma(COAD).By analyzing the relationship between SIK3 expression and immune-infiltrating T cells in the tumor microenvironment,it was found that the SIK3 expression level was positively correlated with the level of cancer-associated fibroblast infiltration in COAD,head and neck squamous cell carcinoma(HNSC),HPV-infected head and neck squamous carcinoma(HNSC-HPV+),pancreatic adenocarcinoma(PAAD),rectal adenocarcinoma(READ),gastric adenocarcinoma(STAD),testicular germ cell tumor(TGCT),and thymic adenocarcinoma(THYM),and that it was negatively correlated with the level of cancer-associated fibroblast infiltration in low-grade gliomas(LGG)of brain.Pan-cancer survival analysis showed that SIK3 high-expression had a poorer prognosis in adrenocortical carcinoma(ACC),and SIK3 low-expression had a poorer prognosis in renal clear cell carcinoma(KIRC),LGG,mesothelioma(MESO),PAAD,and thyroid cancer(THCA).Conclusion:The SIK3 gene plays an important role in the tumor immune microenvironment and has been identified as having prognostic value in a variety of cancers,speculating that the SIK3 gene is a potential target for cancer immunotherapy and tumor biomarkers.
