黄酮类金属配合物抗肿瘤作用的研究进展

摘要 癌症严重威胁着人类的健康与生命,药物治疗已成为当今临床治疗肿瘤的重要手段之一,金属抗肿瘤药物的研究方兴未艾。黄酮类化合物因具有良好配位性质、较强的DNA嵌合能力以及潜在的抗肿瘤活性,成为了金属抗癌药物领域的一大研究热点。研究表明,黄酮在与金属离子配位后,不仅能提升原有的抗肿瘤活性,还有望改善其生物利用度并降低毒副作用。本文根据黄酮类化合物的母体结构特点综述了黄酮类、黄酮醇类、二氢黄酮类、二氢黄酮醇类、异黄酮类以及查耳酮类金属配合物的配位模式及抗肿瘤作用,以期为抗肿瘤类黄酮-金属配合物的研制提供参考依据,助力金属药物在癌症治疗领域中的应用。 Cancer is a major public health problem worldwide,and medication has become one of the most important clinical methods for tumor treatment.Continuous efforts have been made in the investigation of new metallodrugs that are endowed with anticancer property.Flavonoids have drawn considerable attention in the field of metal based anticancer drugs,owing to their excellent coordination properties,DNA binding ability,and potential antitumor bioactivity.Comprehensive studies have demonstrated that the complexation of flavonoids with metals would not only result in the corresponding metallic compounds presenting greater antitumor potency,but also improve their bioavailability and reduce toxic side effects.On this basis,the article intends to review the coordination modes and antitumor properties of the reported flavonoid-metal complexes,according to the characteristics of the mother nucleus structures,such as flavone,flavonol,dihydroflavone,dihydroflavonol,isoflavone,and chalcone,with the aim to provide a theoretical basis for developing related metallic compounds and to promote the application of metallodrugs in the field of cancer therapy.
