circCHST15在透明细胞肾细胞癌中的表达及其与预后的联系

Expression of circCHST15 in clear cell renal cell carcinoma and its association with prognosis

目的探讨circCHST15在透明细胞肾细胞癌(ccRCC)中的表达及其与预后的联系。方法利用qRT-PCR法检测ccRCC组织和匹配的相邻正常组织中circCHST15的表达量;分析ccRCC组织中circCHST15表达量与临床病理特征的相关性;采用Kaplan-Meier曲线分析circCHST15表达与临床预后之间的关联;利用Cox比例风险回归模型进行单因素和多因素回归分析。结果circCHST15在ccRCC组织中的相对表达量为(11.18±4.25),显著高于正常组织的(7.20±2.37)(P=0.009)。circCHST15表达量与TNM分期呈正相关(P<0.01)。Kaplan-Meier曲线分析显示,circCHST15高表达ccRCC患者的总生存期(OS)和无进展生存期(PFS)均短于circCHST15低表达ccRCC患者(P<0.001)。多因素Cox回归分析显示,年龄、TNM分期和circCHST15表达是影响ccRCC患者预后的独立危险因素(P<0.05)。结论circCHST15高表达与ccRCC增殖转移和预后相关。因此,circCHST15可以作为预测ccRCC预后的新型生物标志物。

Objective To investigate the expression of circCHST15 in clear cell renal cell carcinoma(ccRCC)and its association with prognosis.Methods The expression of circCHST15 in ccRCC tissues and matched adjacent normal tissues was detected by qRT-PCR;the relationship between circCHST15 expression and clinicopathological features in ccRCC tissues was analyzed;Kaplan-Meier curve was used to analyze the correlation between circCHST15 expression and clinical prognosis;univariate and multivariate regression analysis was performed by Cox proportional hazard regression model.Results The relative expression of circCHST15 in ccRCC tissues was(11.18±4.25),which was significantly higher than(7.20±2.37)in normal tissues(P=0.009).circCHST15 expression was positively correlated with TNM stage(P<0.01).Kaplan-Meier curve analysis showed that the overall survival(OS)and progression-free survival(PFS)of ccRCC patients with high expression of circCHST15 were shorter than those of the ccRCC patients with low expression of circCHST15(P<0.001).The multivariate Cox regression analysis showed that age,TNM stage and circCHST15 expression were independent risk factors for the prognosis of ccRCC patients(P<0.05).Conclusion The high expression of circCHST15 is associated with proliferation,metastasis and prognosis of ccRCC.Therefore,circCHST15 can serve as a novel biomarker for predicting the prognosis of ccRCC.
