枕神经电刺激对慢性偏头痛模型大鼠疼痛相关行为的影响 被引量:1

Effect of occipital nerve stimulation on pain related behaviors in a rat model of chronic migraine

摘要 目的:探究枕神经电刺激(occipital nerve stimulation,ONS)对慢性偏头痛(chronic migraine,CM)模型大鼠疼痛相关行为的影响。方法:采用炎症汤(inflammatory soup,IS)刺激大鼠硬脑膜模型,采用随机数字表法将雄性SD大鼠分成慢性偏头痛模型大鼠组(CM组)、枕神经电刺激干预慢性偏头痛模型大鼠组(ONS+CM组)、对照组(Con组),每组7只。对CM组和ONS+CM组大鼠连续14天输注IS刺激硬脑膜,每日1次,第15天对ONS+CM组进行ONS,基线及第1、5、9、14次IS输注后24 h及ONS术后24 h测量大鼠头面部多部位机械痛阈及其他疼痛相关行为。机械痛阈采用von Frey测定,其他疼痛相关行为采用CinePlex®行为分析系统,采集大鼠15 min的行为数据。结果:重复IS刺激硬脑膜可使大鼠头面部机械痛阈下降并维持在较低水平,ONS可逆转这种痛阈降低;ONS可逆转因重复IS刺激引起的大鼠水平运动总距离、探索行为及冻结行为的改变。结论:ONS可改善CM模型大鼠痛觉超敏现象,并可逆转部分疼痛相关行为的改变。 Objective:To examine the effect of occipital nerve stimulation(ONS)on pain related behaviors in a rat model of chronic migraine(CM),and to explore the underlying mechanism of ONS in migraine.Methods:Twenty-one male Sprague-Dawley(SD)rats were randomly divided into control(Con)group,inflammatory soup group(CM),inflammatory soup+occipital nerve stimulation group(ONS+CM),7 animals in each group.A classic CM rat model was established by repeated dural infusions of IS.The ONS+CM cohort was implanted bilaterally with electrodes to produce ONS,Periorbital,prefrontal and occipital mechanical threshold were evaluated with von Frey filaments and other pain related behaviors were tested.Results:There is no baseline differences in pain related behaviors among the Con,CM and ONS+CM group.After the CM animal model was established,animal model displayed behavioral results similar to those experienced by migraineurs.Specifically,there was a decrease in routine physical activity and increase in freezing episodes.Also,the animals demonstrated mechanical allodynia in the periorbital,prefrontal and occipital region.They were ameliorated by the treatment of ONS.Conclusion:ONS improves mechanical allodynia and some pain related behaviors in a rat model of CM.
