钛酸钾晶须改性PMMA的生物相容性研究

摘要 目的:评价钛酸钾晶须改性聚甲基丙烯酸甲酯(PMMA)材料的生物相容性。方法:钛酸钾晶须经硅烷偶联剂表面处理后,使用研磨法使之按照一定比例与PMMA粉均匀混合,合成钛酸钾晶须改性PMMA材料。利用高糖培养基分别浸泡PMMA和改性材料制备浸提液作为实验组,原高糖培养基作为阴性对照组。将各组分别与L929细胞共培养,通过噻唑蓝比色法检测细胞增殖,活性氧检测评估细胞氧化损伤,综合评估改性PMMA材料生物相容性。结果:钛酸钾晶须改性PMMA材料组细胞增殖率为85.3%,细胞毒性评级为1级,按照ISO标准认定为无细胞毒性;钛酸钾晶须改性PMMA材料组活性氧检出量显著高于阴性对照组(P<0.001),但与PMMA组无统计学差异。结论:钛酸钾晶须改性PMMA材料具有可接受的生物相容性。 Objective:To evaluate the biocompatibility of PMMA modified by potassium titanate whisker.Methods:Potassium titanate whisker modified PMMA material was prepared by grinding the surface of potassium titanate whisker with silane coupling agent and mixing it with PMMA powder in a certain proportion.PMMA and modified material were soaked in the medium to prepare the leaching liquor as the experimental group,and the original medium as the negative control group.L929 cells were co-cultured with the leaching liquor of each group.The cell proliferation was detected by MTT assay and the oxidative damage was evaluated by reactive oxygen species(ROS)assay,to evaluate the biocompatibility of PMMA modified by potassium titanate whisker.Results:The cell proliferation rate of PMMA modified by potassium titanate whisker was 85.3%,and the cytotoxicity was grade 1,which was considered as non-cytotoxicity according to the ISO standard.The amount of ROS in PMMA modified with potassium titanate whisker was significantly higher than that in the negative control group(P<0.001),but there was no significant difference between the two experimental groups.Conclusion:PMMA modified by potassium titanate whisker have acceptable Biocompatibility.
