新型二氢氧杂蒽结构近红外荧光探针的合成及其在过氧化氢检测中的应用

摘要 目的设计合成新型二氢氧杂蒽(DHX)结构近红外荧光探针并用于过氧化氢(H_(2)O_(2))的原位、痕量、可视化、特异性检测。方法以DHX结构为荧光团,引入吡啶盐作为受体部分构建新型线粒体靶向荧光探针分子DHX-H_(2)O_(2),并对探针的荧光性能、选择性、与H_(2)O_(2)的浓度关系、识别机制进行研究,另外研究了探针在细胞和斑马鱼中H_(2)O_(2)的检测应用。结果探针DHX-H_(2)O_(2)能够以良好的选择性与优异的灵敏度(4.21μmol/L)实现对H_(2)O_(2)的近红外(λem=675 nm)荧光响应。此外,探针具有定位细胞线粒体的性能,皮尔森相关系数为0.97。结论设计合成的探针分子具有选择性好、灵敏度高、细胞毒性低等优点,成功实现了对细胞以及斑马鱼中内源性和外源性H_(2)O_(2)的成像检测,具有广阔的生物应用前景。 Objective To design and synthesize a novel dihydroxanthene(DHX)structure near-infrared fluorescence probe for in situ,trace,visualization,and specific detection of hydrogen peroxide(H_(2)O_(2)).Methods A novel mitochondrial targeted fluorescence probe molecule DHX-H_(2)O_(2)was constructed with DHX structure as fluorophore and pyridine salt as acceptor.The fluorescence properties,selectivity,relationship with H_(2)O_(2)concentration and recognition mechanism of the probe were studied.In addition,the application of the probe in the detection of H_(2)O_(2)in cells and zebrafish were also studied.Results The probe DHX-H_(2)O_(2)could achieve near-infrared(λem=675 nm)fluorescence response to H_(2)O_(2)with good selectivity and excellent sensitivity(4.21μmol/L).In addition,the probe had the ability to locate the mitochondria of cells,with a Pearson correlation coefficient of 0.97.Conclusion The designed and synthesized probe molecule has the advantages of good selectivity,high sensitivity and low cytotoxicity,and has successfully realized the imaging detection of endogenous and exogenous H_(2)O_(2)in cells and zebrafish,which has broad biological application prospects.
