艾绒的HPLC指纹图谱及主要成分含量测定

摘要 目的:建立艾绒饮片的指纹图谱,对其中7种主要成分进行含量测定。方法:采用HPLC法建立指纹图谱,以异绿原酸C为参照,进行相似度评价,确定共有峰,并比对对照品色谱图,测定艾绒中绿原酸、异绿原酸A、隐绿原酸、异泽兰黄素、异绿原酸B、宗矢车菊素、异绿原酸C等7种成分含量。结果:23批艾绒样品确定12个共有峰,指认上述7种成分,线性范围分别为0.0222~0.6660μg、0.0083~0.2490μg、0.0146~0.4380μg、0.0356~1.0680μg、0.3470~1.0410μg、0.0080~0.2400μg、0.0204~0.6120μg(r>0.999);平均加样回收率96.58%~100.85%(RSD为0.47%~1.70%,n=6);含量分别为0.368~2.428 mg/g、0.063~0.810 mg/g、0.148~1.952 mg/g、0.447~4.587 mg/g、0.309~3.407 mg/g、0.227~1.058 mg/g、0.588~2.714 mg/g。结论:该方法专属性强,精密度高,重复性好,为艾绒等级的鉴定提供新的参考依据。 Objective:To establish the fingerprint of Moxa floss,determine the contents of eleven main components and to provide reference for the quality evaluation of Moxa floss.Methods:Fingerprint was established of by HPLC.Using isochlorogenic acid C as reference,HPLC fingerprints of twenty-three batches of Moxa floss were drawn and the similarity evaluation was conducted,the common peaks were also confirmed;and the chromatographic peak was identified by comparing with the chromatogram of reference substance,the contents of seven components as chlorogenic acid,jaceosidin,isochlorogenic acid B,isochlorogenic acid A,epuatilinin,isochlorogenic acid C,cryptochlorogenic acid were determined.Results:There were twelve common peaks in twenty-three batches of Moxa floss,A total of seven common peaks were identified,which were above 7 ingredients.The linear ranges of components were 0.0222-0.6660μg,0.0083-0.2490μg,0.0146-0.4380μg,0.0356-1.0680μg,0.3470-1.0410μg,0.0080-0.2400μg,0.0204-0.6120μg(r>0.999).The average recoveries were 96.58%-100.85%with RSD of 0.47%-1.70%(n=6).The contents were 0.368-2.428 mg/g,0.063-0.810 mg/g,0.148-1.952 mg/g,0.447-4.587 mg/g,0.309-3.407 mg/g,0.227-1.058 mg/g,0.588-2.714 mg/g.Conclusion:Themethod established were specific,high precision and reproducible,which can be used for the quality evaluation and control of Moxa floss.
