基于UHPLC-QE-Orbitrap-MS技术的深加工槟榔籽的成分分析

摘要 本研究以槟榔深加工过程中产生的副产物槟榔籽为研究对象,采用超高效液相色谱-四极杆-静电场轨道肼高分辨质谱法(UHPLC-QE-Orbitrap-MS)对槟榔籽的成分进行分析鉴定。采用ACQUITY UPLC HSS T3色谱柱(2.1 mm×100 mm,1.8μm),以0.1%甲酸水溶液(A)-0.1%甲酸乙腈溶液(B)为流动相进行梯度洗脱,流速0.3 mL·min^(-1),柱温35℃,进样体积10μL,使用电喷雾离子源(ESI),扫描范围m/z为100~1200,Full MSddMS2检测模式,正负离子同时采集。利用Compound Discoverer 3.3软件对原始数据进行分析,通过本地数据库和mzCloud在线数据库对化合物进行结构鉴定。试验鉴定出85种存在于槟榔籽中的成分,分别为生物碱类化合物3种、黄酮类化合物24种、有机酸类化合物15种、酚类化合物7种、萜类化合物7种、氨基酸类化合物6种、糖类化合物4种、酯类化合物4种、酰胺类化合物2种、鞘脂类化合物2种、类固醇衍生物1种、苯丙素类化合物1种、香豆素类化合物1种、其它8种。UHPLC-QE-Orbitrap-MS检测技术可以对槟榔深加工副产物槟榔籽中的成分进行快速识别,本研究为槟榔资源进一步综合利用,开发高附加值产品提供科学依据。 To study the chemical composition of Areca catechu L.seed in deep processing,UH-PLC-QE-Orbitrap-MS technology was used to analyze and identify the composition of Areca cate-chu L.seed.Gradient elution was performed on ACQUITY UPLC HSS T3 column(2.1 mm×100 mm,1.8μm)with 0.1%formic acid aqueous solution(A)to 0.1%formic acid acetonitrile solution(B)as mobile phases.The flow rate was 0.3 mL·min^(-1),the column temperature was 35℃,and the sample volume was 10μL.An electric spray ion source(ESI)was used with a scanning range of 100^(-1)200 m/z and positive and negative ions were collected simultaneously,and the detection mode was Full MS-ddMS2.The original data were analyzed using Compound Discoverer 3.3 software,and the structures of the compounds were identified using the local da-tabase and mzCloud online database.Eighty-five compounds of Areca catechu L.seed in deep processing were detected,including 3 alkaloids,24 flavonoids,15 organic acids,7 phenols,7 ter-penoids,6 amino acids,4 carbohydrates,4 esters,2 amides,2 sphingolipids,1 steroid,1 phe-nylpropanoid,1 coumarin and 8 other species.The UHPLC-QE-Orbitrap-MS detection technolo-gy quickly identified the compound of Areca catechu L.seed in deep processing,and the research results provided scientific basis for exploring the application prospects and developing high value-added products of Areca catechu L.
