小儿感冒宁颗粒对脾虚型功能性消化不良大鼠胃肠运动及血清胃肠激素水平的影响 被引量:1

Influence of Xiaoer Ganmaoning Granule on Gastrointestinal Motility and Serum Gastrointestinal Hormones in Rats with Functional Dyspepsia of Spleen Deficiency

摘要 目的:考察小儿感冒宁颗粒对脾虚型功能性消化不良(FD)大鼠模型的药效作用。方法:特制饲料喂养制备SD大鼠脾虚型FD模型。造模前大鼠随机分为6组,即对照组,模型组,儿宝颗粒组(EB,7.10 g·kg^(-1)·d^(-1)),小儿感冒宁低、中、高剂量组(3.55、7.10、14.20 g·kg^(-1)·d^(-1)),每组10只。除对照组外,其余各组大鼠每日给予特制饲料喂养进行造模。持续造模1周后开始给药,连续20 d。每周2次测定各组大鼠一般状态评分、体质量、24 h进食量、24 h饮水量;给药结束后测定各组大鼠胃排空率、胃重指数、小肠推进率、胸腺指数、脾脏指数;苏木素-伊红(HE)染色法观察大鼠十二指肠、结肠、胃窦部位的组织病理学改变;Image J软件测量小肠绒毛高度、宽度、绒毛间隙、结肠固有层厚度、腺体横截面积、胃窦黏膜浅层破损厚度;酶联免疫吸附法(ELISA)测定血清胃泌素(GAS)、胃动素(MTL)、胆囊收缩素(CCK)、血管活性肠肽(VIP)及瘦素(LEP)含量。结果:与模型组比较,小儿感冒宁颗粒可降低脾虚型FD模型大鼠一般状态评分,缓解体质量增长缓慢及饮水减少,显著增加胃排空率、小肠推进率(P<0.05,P<0.01),改善十二指肠、结肠及胃窦组织病理学特征异常。此外,小儿感冒宁颗粒能够显著升高模型大鼠血清GAS、MTL水平(P<0.05,P<0.01),显著降低血清CCK、VIP、LEP水平(P<0.05,P<0.01)。结论:小儿感冒宁颗粒可通过促进胃排空和小肠推进、调节血清胃肠激素水平起到改善脾虚型FD的作用。 Objective:To investigate the pharmacodynamics of Xiaoer Ganmaoning granule(XEGMN)on the rat model of functional dyspepsia(FD)with spleen deficiency.Methods:The SD rat model of FD with spleen deficiency was established by feeding with a special diet.Before modeling,the rats were randomly divided into six groups:control,model,Erbao(EB,7.10 g·kg^(−1)·d^(−1)),XEGMN-D,XEGMN-Z,and XEGMN-G(3.55,7.10,and 14.20 g·kg^(−1)·d^(−1))groups,with 10 rats in each group.Except for the normal group,the other groups were fed with a special diet daily to establish the model.After one week of continuous modeling,the rats were given drugs for 20 days.The general status score,body weight,24 h food intake,and 24 h water intake were measured twice a week.After the treatment,the gastric emptying rate,gastric weight index,small intestinal propulsion rate,thymus index,and spleen index were measured.The histopathological changes in the duodenum,colon,and antrum of rats were observed by hematoxylin-eosin(HE)staining.The height,width,villous space of intestinal villi,the thickness of lamina propria of the colon,the cross-sectional area of glands,and the thickness of superficial damage of gastric antrum mucosa were measured by the Image J software.The serum levels of gastrin(GAS),motilin(MTL),cholecystokinin(CCK),vasoactive intestinal peptide(VIP),and leptin(LEP)were measured by ELISA.Results:Compared with the model group,Xiaoerganmaoning granule could reduce the general status score of FD model rats with spleen deficiency,relieve the slow weight gain,decrease the water intake,significantly increase the gastric emptying rate and small intestinal propulsion rate(P<0.05,P<0.01),and improve the abnormal histopathological features of duodenum,colon,and gastric antrum of the model rats.In addition,Xiaoerganmaoning granule could significantly increase the levels of GAS and MTL in serum(P<0.05,P<0.01)and significantly reduce the contents of CCK,VIP,and LEP in serum(P<0.05,P<0.01)in the model rats.Conclusion:Xiaoerganmaoning granule can improve FD with spleen deficiency by promoting gastric emptying and small intestine propulsion and regulating serum gastrointestinal hormone levels.
